摘要
目的研究慢性社会击败应激所致抑郁样小鼠肠黏膜屏障功能性变化。方法构建慢性社会击败应激小鼠模型,运用社会接触实验检测小鼠抑郁样行为,将小鼠分为抑郁敏感组(10只)、抑郁抵抗组(6只)、空白对照组(10只),HE染色制片观察肠上皮的组织形态学变化并评分,实时荧光定量PCR测定Muc2基因的表达量。结果与空白对照组相比,抑郁敏感组小鼠回肠组织形态结构出现明显损伤,且组织病理学评分显著升高,差异具有统计学意义(P<0.05),抑郁抵抗组与空白对照组差异不显著(P>0.05);抑郁敏感组M uc 2的mRNA表达上调,差异具有统计学意义(P<0.05),抑郁抵抗组差异不显著(P>0.05)。结论慢性社会击败应激所致小鼠抑郁症可能与肠黏膜机械屏障损伤存在相关性。
Objective To study functional changes of intestinal mucosal barrier in depressed mice induced by chronic social defeated stress.Methods The chronic social defeat stress mice model was used with the social interaction test to determine depression behavior in mice.According to the result of the test,mice were divided into sensitive depression group,resistive depression group and blank control group.The morphological changes of intestinal epithelial tissue and grading were observed by HE staining.Using the real time fluorescence quantitative PCR assay,the expression of the Muc2 gene were measured.Results HE staining showed that compared with the blank control group,the morphological structure of ileum in mice with sensitive depression showed obvious injury,and the histopathological score was significantly increased.Real-time fluorescence quantitative PCR showed that the mRNA expression of Muc 2 mRNA was up-regulated in the sensitive depression group compared with the blank control group.Conclusion Chronic social frustration stress may have a correlation with intestinal mucosal mechanical barrier injury in mice with depression.
