通过集成CRISPR/Cas9、受精卵注射和核移植技术快速高效制备胰腺缺陷小型猪模型

Production of Pancreatic Defect Bama Miniature Pig Model with the Combination of CRISPR/Cas9,Zygote Injection and Somatic Cell Nuclear Transfer Technologies

摘要 小型猪在体型大小,生理特点上与人类具有的很大的相似性,因此愈来愈多的应用于人类疾病模型及再生医学相关模型.多能性细胞的分化潜能赋予了其器官再生的能力,继大小鼠器官异种再造后人类器官在猪模型中的制备已经成为近几年的研究热点.为了得到胰腺缺陷小型猪,本研究应用CRISPR/Cas9技术,受精卵胞质注射技术联合核移植技术快速高效地获得了基因型相同的胰腺缺陷的小型猪模型,为小型猪疾病模型的制备,人类器官异体再造提供了借鉴意义. Bama miniature pig share similar physiological features with human,as well as the weight and size of organs,which provide us opportunities to produce valuable models for human disease and regenerative medicine researches.The differentiation potential of tissue-specific pluripotent cells enable the related organs to regenerate.Followed the heterogeneous reconstruction of mouse organs in rats,the study of the reconstruction of human organs in pig models emerged and became the research hotspot in recent years.In order to obtain the miniature pig pancreas defect model,CRISPR/Cas9,zygote cytoplasmic injection and somatic cell nuclear transfer technologies were used to generate pancreas defect miniature pig model with same genotype at high efficiency.This study provided useful reference for miniature pig disease model creation and human organ heterogeneous reconstruction researches.
