人多能干细胞体外T细胞分化的研究进展

Research Progress of T Cell Oriented Differentiation of Human Pluripotent Stem Cells in vitro

T淋巴细胞是免疫系统中的重要组分,在临床上可用于免疫治疗.尽管T细胞可以从患者外周血中分离和扩增,但是在数量上还是有一定限制,而且对T细胞进行基因编辑的困难度要大于多能干细胞.而具有无限增殖特性的人多能干细胞可以提供源源不断的T细胞,对人多能干细胞进行基因编辑还可获得"通用型"(Universal)T细胞治疗产品.所以研究人多能干细胞分化为成熟的T细胞意义重大.本文主要介绍人多能干细胞体外分化获得T细胞的理论基础和最新进展.

As an important component of the immune system,T lymphocytes could be used in clinical immunotherapy.T cells could be isolated and proliferated from patient’s peripheral blood,however,the number is limited and they are more difficult to conduct gene editing than pluripotent stem cells.Human pluripotent stem cells,which have unlimited proliferation capability,can provide an endless source of T cells.Gene editing of human pluripotent stem cells can also lead to the production of " Universal" T cell for cell therapy.Thus,researches on the differentiation of pluripotent stem cells into mature T cells is of great significance.This paper introduced the theoretical basis and reviewed recent research progress of T cell derived from human pluripotent stem cells.
