双扩增法和real-time RT-PCR法检测手足口病的比较

Comparative analysis of dual amplification and real-time RT-PCR for hand-foot-mouth disease

目的开展双扩增法和real-time RT-PCR法检测手足口病的比较性研究,为基层实验室建立手足口病科学快速检测方法提供依据。方法对2016年-2017年宜昌市9县市区采集的205份手足口病患者的咽拭子样本,同时采用双扩增法和real-time RT-PCR法检测手足口病通用病毒、CoxA16、EV71,比较2种方法检测结果的差异,对2种方法检测结果相异的标本进行测序。结果双扩增法同real-time RT-PCR法检测结果比较,阳性标本数差异无统计学意义(x^2=0.743,P>0.05);且2种方法与测序方法结果比较,符合数差异无统计学意义(x^2=3.32,P>0.05)。结论双扩增法适合基层实验室推广使用。

Objective Comparative study was carried out on the detection of hand-foot-mouth disease(HFMD)by double amplification and real-time RT-PCR,so as to provide basis for rapid detection in primary Laboratory.Methods 205 throat swabs of hand,foot,mouth disease cases which were collected from 9 counties of Yichang City during 2016-2017 were simultaneously conducted for detection of universal enterovirus,Cox A16 and EV71 by dual amplification and real-time RT-PCR,and the results were compared.DNA sequencing was conducted for samples which had inconsistent results in the two methods.Results There was no statistical significance on the difference in the number of positive specimens between dual amplification and real-time RT-PCR(x^2=0.743,P>0.05).And there was no statistical significance on the difference of consistence number between the two methods and sequencing methods(x^2=3.32,P>0.05).Conclusion The dual amplification technique is suitable for the promotion and application in primary laboratories.
