五味子酸枣仁软胶囊改善动物睡眠功能的研究

Study on Improving the Sleep Function of Animals Through Treatment of Schisandra chinensis Suanzaoren Soft Capsule

主要探究五味子酸枣仁软胶囊对改善动物睡眠功能的影响。选取SPF级KM雌性小鼠48只(体重18.00~22.00 g)。试验设4组,包括高、中、低3个剂量组和1个阴性对照组,高剂量组为1.00 g/kg(相当于成人每日用量的30倍)、中剂量组为0.33 g/kg(相当于成人每日用量的10倍)、低剂量组为0.17 g/kg(相当于成人每日用量的5倍)、阴性对照组为玉米胚芽油。小鼠每日灌胃给予受试样品1次,灌胃用量20 mL/kg,连续30 d。通过直接睡眠试验、延长戊巴比妥钠睡眠时间试验、戊巴比妥钠阈下剂量睡眠试验、巴比妥钠睡眠潜伏期试验,探究该样品对改善动物睡眠功能的影响。结果表明,在该试验条件下,延长戊巴比妥钠睡眠时间试验、戊巴比妥钠阈下计量睡眠时间试验、巴比妥钠睡眠潜伏期3项试验中两项呈阳性,且无明显直接睡眠作用。根据《保健食品检验与评价技术规范》中改善睡眠功能检验方法,可判定该样品具有改善睡眠功能。

This experiment mainly explored the effect of Schisandra Chinensis Suanzaoren Soft Capsule on improving sleep function of animals.Forty-eight female SPF KMmice(weighing 18.00-22.00 g)were selected.Four groups were set up,including high(1.00 g/kg,i.e.30 times the daily dosage of human),medium(0.33 g/kg,i.e.10 times the daily dosage of human)and low(0.17 g/kg,i.e.5 times the daily dosage of human)dose groups and a negative control group(corn germ oil).The mice were given the sample once a day by gavage with a volume of 20 mL/kg for 30 days.The effects of this sample on sleep function of animals were investigated by direct sleep experiment,prolonged pentobarbital sodiumsleep time experiment,pentobarbital sodiumsubthreshold dose sleep experiment and barbital sodiumsleep latency experiment.The results showed that two of the three experiments were positive in prolonging the sleep time of sodiumpentobarbital,subthreshold measurement of sleep time of sodiumpentobarbital and the sleep latency of sodiumbarbital,and there was no obvious direct sleep effect.According to the test method of improving sleep function in Technical Specification for Inspection and Evaluation of Health Food,we conclude that the sample improvessleep function.
