云上黑山羊MSTN基因的表达差异与发育性变化研究

Research on Differences and Development Changes of MSTN Gene Expression in Yunshang Black Goat

为研究云上黑山羊肌肉组织中肌肉生长抑制素(Myostatin,MSTN)基因mRNA表达发育性变化规律,应用实时荧光定量PCR技术分析了MSTN基因在3、6、10和12月龄云上黑山羊腰肌、背最长肌和腿肌中的相对表达量,以期为云上黑山羊肉用性状的选育提供可借鉴的理论资料。结果显示:云上黑山羊4个生长阶段,MSTN基因在肌肉中的表达模式不尽相同,3月龄和10月龄的表达模式为背肌>腰肌>腿肌,6月龄为腰肌>背肌>腿肌,12月龄为腰肌>腿肌>背肌;3种肌肉组织均在6月龄时表达量最高,不同生长阶段肌肉中MSTN基因的表达均没有出现随着日龄的增加一直增加或下降的趋势,整体表现出先上升后下降的趋势。说明MSTN基因在不同部位的肌肉组织中具有不同的表达模式,且不同生长阶段表达量存在差异。

In order to research the expression law of MSTN gene in the muscle of Yunshang Black goat,the mRNA relative expression in the psoas,longissimus dorsi and leg muscle of Yunshang Black goat from 3,6,10 and 12 months were analyzed by SYBR Green real-time PCR technique,which provided theoretical data for breeding meat trait of Yunshang Black goat.The results showed that the expression pattern of MSTN gene were different in the four growth stages.The expression pattern at 3 months and 10 months were longissimus dorsi>psoas>leg muscle,the expression pattern at 6 months was psoas>longissimus dorsi>leg muscle and at 12 months was psoas>leg muscle>longissimus dorsi;the expression levels of three kinds muscles were the highest at 6 months,the expression of MSTN gene in muscles at different growth stages did not increase or decrease with the increase of day age,and the overall expression showed a tendency of first increasing and then decreasing.MSTN gene had different expression patterns in different parts of muscle tissue,and the expression levels were different in different growth stages.
