摘要
以黑杨派的无性系花粉为试验材料,测试其在4℃冰箱储存50天后的花粉活力,目的是为黑杨派杂交育种的父本花粉选择提供依据。结果表明,黑杨派无性系花粉活力测定适宜的培养基成分包括1%的琼脂、15%的蔗糖和10mg/L的硼酸,花粉培养26h时萌发率最高。黑杨派无性系的花粉在4℃冰箱储存50天后,花粉萌发率存在较大变异,其变异范围为0~32%,北方型美洲黑杨、南方型美洲黑杨和欧洲黑杨的花粉活力最高的无性系分别是T144、L23和欧黑5号,其花粉萌发率分别是29.7%、32.0%和11.3%。
With the pollen of Aigeiros clones as test material,the pollen vitality was tested after the pollen was reserved in 4℃refrigerator for 50 days.The aim was to provide a basis for pollen selection of male parent in cross breeding.The results showed that the suitable medium components included 1%agar,15%sucrose and 10 mg/L boric acid,and the germination rate was the highest when the pollen was cultured for 26h.After the pollen reserved in 4℃refrigerator for 50 days,there were large variations in pollen germination rate,the variation range of 0-32%.The clones T144,L23 and Ouhei 5 had the highest pollen vitality among the north type clones of Populus deltoides,the south type clones of Populusdeltoides and the clones of Populus nigra,and their pollen germination rate is 29.7%,32.0%and 11.3%respectively.
