摘要
为了探讨多溴联苯醚-47(PBDE-47)对背角无齿蚌(Anodonta woodiana)慢性毒性效应,克隆了背角无齿蚌热休克蛋白基因(AwHSP60),分析基因全长、开放阅读框、编码、氨基酸。研究发现AwHSP60具有HSP60家族的标签序列;荧光定量PCR分析表达显示,AwHSP60基因广泛分布于斧足、鳃、肝胰脏、闭壳肌、心脏、血淋巴和外套膜,PBDE-47处理后肝胰脏中AwHSP60 mRNA水平在第1-15天显著性增加了89.9%,鳃中AwHSP60 mRNA水平显著性增加了2.09倍,血淋巴中AwHSP60 mRNA水平显著性增加2.13倍。结果表明,背角无齿蚌上调AwHSP60表达有助于增强动物对PBDE-47处理的耐受能力。
In order to investigate the chronic effect of polybrominated diphenyl ether-47(PBDE-47),one heat shock protein 60 was cloned and named AwHSP60.Molecular characterizations of AwHSP60 were analyzed.AwHSP60 was widely distributed in foot,gill,hepatopancreas,adductor muscle,heart,hemocytes and mantle.Administration of PBDE-47 could result in a significant up-regulation of AwHSP60 expression in the hepatopancreas,gill and hemocytes.In the hepatopancreas,compared with that of control group,mRNA level of AwHSP60 increased more than 89.9%from day 1 to 15.In the gill,during experiment observed,expression of AwHSP60 increased more 2.09 times in contrasted with that of control group.In the hemocytes,AwHSP60 expression of PBDE-47 treated group increased more 2.09 times and 1.81 times compared with that of control group.These results indicated that up-regulation of AwHSP60 expression is contribute to enhancing adaption of bivalve A.Woodiana exposed to PBDE-47 treatment.
