摘要
通过对50个香菇(Lentinula edodes)栽培菌株的β1-微管蛋白部分基因进行测序、聚类和构建多态性图谱。结果表明:含有3条不同序列的菌株数为1,含有2条不同序列的菌株数为37,含有1条序列的菌株数为12;89条序列分为3个类型:Ⅰ型包括66条序列,Ⅱ型包括12条序列,Ⅲ型包括9条序列;根据对基因序列的分型,30个菌株属于Ⅰ型,11个菌株属于Ⅰ和Ⅱ型,5个菌株属于Ⅰ和Ⅲ型,1个菌株属于Ⅱ和Ⅲ型,1个菌株属于Ⅲ型;Cr02属于Ⅰ、Ⅲ和待定型,广香9号属于Ⅰ和待定型。
Partial β1-tubulin genes of 50 Lentinula edodes cultivars were sequenced for multiple clones,clustered for their genotypes,and then analyzed for their polymorphism.The results showed that 1 cultivar contained three different β1-tubulin sequences,37 cultivars contained two different sequences,and twelve cultivars contained a single sequence.These partial β1-tubulin sequences encoded an identical amino acid sequence and were grouped into genotypeⅠ,Ⅱ and Ⅲ by clustering analysis,respectively.Type Ⅰ,Ⅱ and Ⅲ group had 66,12 and 9 sequences,respectively.According to the genetic typing result,30 cultivars contained a type Ⅰ β1-tubulin gene;11 cultivars contained both type Ⅰ and Ⅱ β1-tubulin genes;5 cultivars contained both type Ⅰ and Ⅲ β1-tubulin genes;1 cultivar contained both Ⅱ and Ⅲ β1-tubulin genes;1 cultivar contained a type Ⅲ β1-tubulin gene;cultivar Cr02 contained type Ⅰ,Ⅲ and an undefined β1-tubulin gene;and cultivar Guangxiang No.9 contained type Ⅰ and an undefinedβ1-tubulin gene.These results provided theoretical support for identifying L.edodes cultivars in China.
