红缘拟层孔菌子实体提取物的抗炎镇痛解热活性研究 被引量:4

Study on Anti-inflammatory, Analgesic and Antipyretic Activities of Extracts of Fomitopsis pinicola Fruiting Bodies

摘要 利用二甲苯致小鼠耳肿胀和醋酸致小鼠腹腔毛细血管通透性实验,研究红缘拟层孔菌(Fomitopsis pinicola)子实体不同提取物的抗炎作用,通过测定小鼠血清超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)水平,炎症组织中SOD、MDA、前列腺素E 2(prostaglandin E 2,PGE 2)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平探究其抗炎机理;运用醋酸致小鼠扭体和热板致小鼠疼痛方法研究其镇痛作用;利用干酵母致大鼠发热和内毒素致大鼠发热模型探究其解热活性。结果表明,与模型组相比,红缘拟层孔菌子实体极性高的水提物和正丁醇提取物能显著降低腹腔毛细血管通透性、小鼠醋酸扭体和热板疼痛反应、干酵母和内毒素致大鼠发热的体温,其次为甲醇提取物;而二氯甲烷和石油醚提取物可显著抑制二甲苯导致的小鼠耳肿胀,显著升高小鼠血浆和右耳组织液中SOD水平,并显著降低小鼠血清MDA和右耳组织液MDA和PGE 2水平,说明红缘拟层孔菌子实体提取物具有抗炎、镇痛和解热作用,且其活性物质主要集中在极性高的水和正丁醇提取物中,其次为甲醇提取物中。 Anti-inflammatory effects of different extracts of Fomitopsis pinicola fruiting bodies were investigated by xylene-induced ear swelling test and acetic acid-induced peritoneal capillary permeability test in mice.The anti-inflammatory mechanisms were studied via determination of serum levels of superoxide dismutase(SOD)and Malondialdehyde(MDA),as well as tissue levels of SOD,MDA,prostaglandin E 2(PGE 2)and tumor necrosis factor-α(TNF-α).The analgesic effects were assessed by acetic acid-induced writhing and hot plate-induced pain in mice.The antipyretic activities were explored using dry yeast-induced rat fever and endotoxin-induced rat fever models.The results showed that,compared with the model group,water extract and n-butanol extract(high polarity extracts)of F.pinicola significantly reduced peritoneal capillary permeability,acetic acid-induced writhing and hot plate pain response in mice,and the body temperature in rat fever models induced by yeast or endotoxin(P<0.01 or P<0.05),followed by methanol extract(P<0.05).Methylene chloride and petroleum ether extracts significantly reduced xylene-induced ear swelling in mice(P<0.01),and significantly increased SOD levels in mouse sera and right ear tissues(P<0.01 or P<0.05),and significantly decreased the levels of MDA in mouse sera and MDA and PGE 2 in right ear tissues(P<0.01 or P<0.05).The results indicated the anti-inflammatory,analgesic and antipyretic effects of F.pinicola fruiting body extracts.The bioactive compounds were mainly concentrated in the highly polar water and n-butanol extracts,followed by methanol extracts.
