绣球菌酸性多糖的分离纯化、结构鉴定及抗氧化活性研究 被引量:16

摘要 采用超声波辅助水提醇沉法提取绣球菌(Sparasis crispa)多糖,并通过HZ-830大孔树脂、DEAE-52纤维素及Sephadex G-100凝胶纯化,收集绣球菌多糖SCP-Ⅱ;采用苯酚硫酸法和间羟基联苯法分别测定SCP-Ⅱ中总糖和糖醛酸含量;采用紫外-可见光谱、高效凝胶渗透色谱、离子色谱、红外光谱、扫描电镜和原子力显微镜对其结构进行鉴定;通过测定总还原力、清除DPPH、OH和O-2自由基的能力来研究其体外抗氧化活性。结果表明:SCP-Ⅱ的总糖、糖醛酸含量分别为98.7%、17.8%,相对分子质量为5.8×10^3,单糖组成为半乳糖、葡萄糖、木糖、果糖(摩尔比为2∶7∶1∶2);其具有一定的清除自由基能力,清除DPPH、OH、O2^-自由基的IC 50分别为1.64、0.94 mg/mL和7.25 mg/mL。 Acidic polysaccharide SCP-Ⅱin Sparassis crispa was first extracted by ultrasonic-assisted water extraction and alcohol precipitation,and then purified by HZ-830 macroporous resin,DEAE-52 cellulose and Sephadex G-100 gel.The content of total sugar and uronic acid in SCP-Ⅱwere determined by the phenol sulfate method and m-hydroxybiphenyl method,respectively.Ultraviolet-visible spectrum,high performance gel permeation chromatography,ion chromatography,infrared spectrum,scanning electron microscopy and atomic force microscopy were used to identify the structure of SCP-Ⅱ.The antioxidant activity of SCP-Ⅱin vitro was investigated through measuring the total reduction ability and ability to scavenge free radicals(DPPH,OH,and O2^-).The results showed that SCP-Ⅱhad a molecular weight of 5.8×10^ 3.Its total sugar and uronic acid content were 98.7%and 17.8%,respectively.SCP-Ⅱcomprised galactose,glucose,xylose and fructose at a molar ratio of 2∶7∶1∶2.It had certain free radical scavenging ability and total reduction ability.The IC 50 of DPPH,OH and O-2 free radicals were 1.64,0.94 mg/mL and 7.25 mg/mL,respectively.
