摘要
目的优化秦皮多酚超声提取工艺条件,评价其抗氧化活性。方法以秦皮多酚提取率为考察指标,乙醇体积分数、超声时间、超声功率及料液比为影响因素,进行单因素实验,在此基础上采用响应面法优化秦皮多酚的提取工艺;选取维生素C作为阳性对照,考察秦皮多酚对1,1-二苯基-2-三硝基苯肼(DPPH)及2,2′-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基的清除能力,评价其体外抗氧化活性。结果秦皮多酚的最佳超声提取工艺参数为体积分数为45%的乙醇,超声时间为33 min,超声功率为90 W,料液比为1∶26,秦皮多酚的提取率预测值为6.232%。秦皮多酚和维生素C抑制DPPH自由基的IC 50分别为6.217和6.032μg·mL^-1;抑制ABTS自由基的IC 50分别为4.048和3.639μg·mL^-1。结论采用响应面法优选获得的秦皮多酚提取工艺稳定、可行。秦皮多酚具有一定的体外抗氧化活性。
Objective To optimize the extraction technology of polyphenols from Fraxini Cortex and evaluate its antioxidant activity in vitro.Methods By using the extraction amount of polyphenols from Fraxini Cortex as response value,ethanol volume fraction,solid-liquid ratio,extraction time and extraction power as response factors,response surface method was used to optimize the extraction technology of polyphenols from Fraxini Cortex based on single factor test.By using vitamin C as a positive control,scavenging on 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radicals and 2,2′-azino-bis(3-ethyl benzothiazoline-6-sulfonic acid(ABTS)free activities of polyphenols from Fraxini Cortex were investigated.And its antioxidant activity in vitro was evaluated.Results The optimized extraction conditions for polyphenols from Fraxini Cortex was as follows:ethanol volume fraction of 45%with extraction time 33 min,extraction power 90 W and liquid-solid ratio 1∶26.The extraction amount of polyphenols from Fraxini Cortex was 6.232%in the verification test.The inhibition of DPPH free radical IC 50 were 6.217 and 6.032μg·mL^-1 respectively by Fraxini Cortex polyphenols and vitamin C;and the inhibition of ABTS free radical IC 50 were 4.048 and 3.639μg·mL^-1 respectively by Fraxini Cortex polyphenols and vitamin C.Conclusion The extraction technology of polyphenols from Fraxini Cortex optimized by response surface method is stable and feasible.The polyphenols from Fraxini Cortex shows certain antioxidant activity in vitro.
