HPLC-电化学法在血清利培酮及9-羟利培酮检测中的应用 被引量:5

摘要 目的建立检测血清利培酮和9-羟利培酮水平的高效液相色谱(HPLC)-电化学法并进行性能验证。方法将高效液相色谱仪与基尔电化学检测器联用。基尔电化学检测器检测电压为1 700 mV,检测电流为0.1~1μA,采用InertSustainSwift C18色谱柱,色谱柱温度为40℃,流速1 mL/min,流动相为辛烷磺酸钠+磷酸+水混合液∶乙腈=73∶27(V∶V)。以叔丁基甲醚为萃取剂。对建立的HPLC-电化学法进行方法学评价。结果HPLC-电化学法检测9-羟利培酮、利培酮和内标的保留时间分别为18.5、8.1及7.5 min,在2.5~100.0 ng/mL范围内线性良好。利培酮低、中、高浓度的日内回收率分别为100.30%、92.25%、97.44%,日内相对标准偏差(RSD)分别为4.18%、6.40%、5.32%;日间回收率分别为98.90%、100.85%、99.64%,日间RSD分别为6.88%、8.87%、6.54%;9-羟利培酮低、中、高浓度的日内回收率分别为96.90%、100.35%、98.04%,日内RSD分别为4.85%、6.08%、5.67%,日间RSD分别为5.06%、7.79%、6.08%。反复冻融3次、室温放置48 h、室外烈日暴晒2 d对利培酮和9-羟利培酮的检测结果无明显影响。利培酮和9-羟利培酮检测限均为0.1 ng/mL,定量限均为0.8 ng/mL。临床常用药物氯氮平、盐酸氯丙嗪、奥氮平、地西泮、富马酸奎硫平、阿立派唑、盐酸齐拉西酮对利培酮、9-羟利培酮的检测不存在干扰。采用HPLC-电化学法和毛细管电泳(CE)-质谱(MS)法同时检测6例单服利培酮的患者的血清样本,2种方法的变异系数(CV)<10%。结论建立的HPLC-电化学法的方法学性能符合临床要求,适用于利培酮和9-羟利培酮的常规检测。
