甘草中小分子水仙苷对溶菌酶淀粉样聚集的影响

摘要 淀粉样蛋白聚集抑制剂的开发对蛋白构象病的防治具有重要意义。文章将溶菌酶作为淀粉样蛋白模型蛋白,通过ThT荧光实验、原子力显微镜、圆二色光谱和分子对接等方法,研究了甘草中的小分子水仙苷对溶菌酶淀粉样聚集的影响。结果表明,水仙苷能有效抑制溶菌酶的淀粉样聚集,改变其聚集路径,使之产生以α-螺旋构象为主的聚集体,有利于缓解细胞毒性。此外,Cu^2+会促使溶菌酶形成以β-折叠构象为主的有毒聚集体,而水仙苷对这一作用也有较好的抑制效果。 The development of amyloid aggregation inhibitors is of great significance for the prevention and treatment of protein conformational diseases. In this work, lysozyme was used as a model protein, and the inhibiting effect of narcissin on its aggregation was studied via ThT fluorescence experiment, atomic force microscopy, circular dichroism spectroscopy and molecular docking. The results showed that narcissin could effectively inhibit the amyloid aggregation of lysozyme and change its aggregation pathway to produce aggregates mainly composed of α-helical conformation, which is beneficial to alleviate cytotoxicity. In addition, it was found that Cu2+ promoted the formation of toxic lysozyme aggregates which are rich in β-sheet conformation, while narcissin well inhibited this effect.
