聚合酶链式反应法和VITEK-2 compact仪器法检测耐甲氧西林凝固酶阴性葡萄球菌的比较 被引量:2

摘要 目的:应用mecA基因聚合酶链式反应(PCR)法和VITEK-2 compact仪器法检测耐甲氧西林凝固酶阴性葡萄球菌,比较仪器法表型检测与PCR法基因型检测的符合性。方法:对97株凝固酶阴性葡萄球菌菌株进行耐药基因mecA扩增,与全自动细菌鉴定及药敏分析系统VITEK-2 compact的结果进行比较。结果:VITEK-2 compact仪器法共检测出76株耐甲氧西林凝固酶阴性葡萄球菌,与mecA基因PCR法的符合率为97.4%(76/78)。2株PCR法检测mecA基因阳性的凝固酶阴性葡萄球菌对苯唑西林敏感。结论:VITEK-2 compact仪器法可以快速、准确地鉴定耐甲氧西林凝固酶阴性葡萄球菌,与PCR法基因型检测符合性高。 Objective:Polymerase chain reaction(PCR)and VITEK-2 compact were used to detect methicillin-resistant coagulase-negative staphylococcus.The coincidence between phenotype detection by instrument and genotype detection by PCR was compared.Methods:97 coagulase-negative staphylococcus strains were amplified by mecA,and compared with the results of VITEK-2 compact,an automatic bacterial identification and drug sensitivity analysis system.Results:76 methicillin-resistant coagulase-negative staphylococci were detected by VITEK-2 compact instrument.The coincidence rate with mecA gene PCR was 97.4%(76/78).Two strains of coagulase-negative staphylococci with mecA gene positive were detected by PCR to be susceptible to oxacillin.Conclusion:VITEK-2 compact can quickly and accurately identify methicillinresistant coagulase-negative staphylococci,which is highly consistent with the genotype detection by PCR.
