蜀葵子生药学检测与芦丁、山奈素和槲皮素的含量测定 被引量:6

摘要 依据《中国药典》(2015版)对蜀葵子进行生药学检测,并新建了蜀葵子中芦丁、山奈素和槲皮素的薄层色谱和高效液相色谱检测方法。结果表明,对比10批蜀葵子药材,其横切面有木栓层细胞数列,4~7束维管束排列成环带,外被大量非腺毛;10批蜀葵子中芦丁、山奈素和槲皮素的R f值均与标准品一致,且斑点清晰。蜀葵子中芦丁、山奈素和槲皮素的平均含量分别为0.617 mg·g^-1、0.034 mg·g^-1和0.033 mg·g^-1;线性范围分别为2.55~206.67μg·mL^-1(R=0.9993)、2.38~199.33μg·mL^-1(R=0.9994)和2.88~233.33μg·mL^-1(R=0.9997);精密度RSD(n=6)分别为0.263%、0.290%、0.405%;平均加标回收率分别为99.86%(RSD=1.83%)、100.72%(RSD=2.03%)和105.91%(RSD=1.87%)。为蜀葵子药材质量控制标准提升奠定了基础。 We studied the pharmacognostic characters of hollyhock seed according to the methods in Chinese Pharmacopoeia(edition 2015),and developed TLC and HPLC methods for the determination of rutin,kaempferol,and quercetin of hollyhock seed.The results show that comparison between ten batches of hollyhock seeds reveals there exist cork cells series on transverse,4-7 vascular bundles arrange in annular shape,and nonglandular hairs in the slice of hollyhock seed.Moreover,the R f values of rutin,kaempferol,and quercetin in ten batches of hollyhock seed are consistent with those of standards,even TLC spots are clear.Furthermore,the average contents of rutin,kaempferol,and quercetin of hollyhock seed are 0.617 mg·g^-1,0.034 mg·g^-1,and 0.033 mg·g^-1,respectively,and the linear ranges are 2.55-206.67μg·mL^-1(R=0.9993),2.38^-199.33μg·mL^-1(R=0.9994),and 2.88-233.33μg·mL^-1(R=0.9997),respectively.The RSDs(n=6)of precision experiment are 0.263%,0.290%,and 0.405%,respectively,while the average adding standard recoveries are 99.86%(RSD=1.83%),100.72%(RSD=2.03%),and 105.91%(RSD=1.87%).The study lays the foundation for the quality standard improvement of hollyhock seed.
