摘要
试验旨在研究MSTN基因启动子区多态性与黄杂鸡体重和体尺指标间的相关性,探寻可用于黄杂鸡选育的分子遗传标记。从乐山市井研养鸡场随机选取92只黄杂鸡,其中母鸡58只,公鸡34只,分别对其体重和各体尺指标进行测定,同时采用PCR-RFLP方法检测MSTN基因启动子区的单核苷酸多态性(SNP)。结果表明,在ATG上游1782位(A→G)发现1个SNP位点,即SNP(c.-1782A>G)。分析MSTN基因多态性与体重和体尺指标的关系,表现为三种基因型:CC、CT和TT,CC基因型的个体在各项指标中显著高于CT和TT基因型个体。基因型与体重和体尺指标的相关性分析结果表明,CC基因型的黄杂鸡体斜长极显著高于CT和TT基因型黄杂鸡的(P<0.01),CC基因型的个体体重显著高于CT和TT基因型黄杂鸡(P<0.05),其余黄杂鸡指标性状在不同基因型组间差异不显著(P>0.05)。说明MSTN基因启动子区SNP(c.-1782A>G)与黄杂鸡的体重和体斜长密切相关,该SNP可以作为黄杂鸡选育的分子遗传标记之一。
In order to study the correlation between the polymorphism of the MSTN gene promoter and the body weight of the yellow hybrid chicken,and explore the molecular genetic markers that can be used in the breeding of yellow hybrid chickens,a total of 92 yellow hybrid chickens are randomly selected from the chicken farm in Jingyan County,Leshan,including 58 hens and 34 cocks.The body weight and body size indicators are measured,and the single nucleotide polymorphism(SNP)of the promoter region of MSTN gene is detected by PCR-RFLP,and the correlation between the polymorphism and the body weight and the body size of the yellow hybrid chicken is analyzed.The results show that there is a SNP site(c.-1782A>G)in the promoter region of the MSTN gene.There are three genotypes detected:CC,CT,and TT,of which CC genotype individuals are higher in each index than individuals with CT and TT genotypes.The analysis of the correlation between genotypes and body weight and body size indicators shows that CC genotype has a significantly greater slanting length than CT and TT genotypes(P<0.01),that CC genotype is significantly heavier than CT and TT genotypes(P<0.05)and that there is no obvious difference in other indicators of different genotypes(P>0.05).Thus,it is clear that SNP(c.-1782A>G),MSTN gene promoter polymorphism,is closely related to the body weight and slanting length of yellow hybrid chickens,and the SNP can be used as one of the molecular genetic markers in the breeding of yellow hybrid chickens.
