聚类分析和主成分分析法研究檀香HPLC特征图谱 被引量:7

摘要 目的:建立檀香药材甲醇提取部位的HPLC特征图谱,为合理地评价檀香药材质量提供方法与依据。方法:采用Waters Symmetry C18色谱柱,以乙腈-0.2%磷酸溶液为流动相,梯度洗脱;检测波长为205 nm,柱温为30℃,流速为1.0 mL/min,建立13批檀香样品的HPLC特征图谱,并进行相似度评价、聚类分析和主成分分析;同时测定样品中挥发油含量。结果:建立了13批檀香样品的特征图谱,共标记8个共有峰,相似度为0.369~0.993,差异较大;聚类分析和主成分分析结果一致;13批檀香样品挥发油含量为0.58%~5.52%,其中S3、S4号样品挥发油含量偏低,与相似度评价结果一致。结论:本研究建立的檀香药材HPLC特征图谱检测方法简便快速,专属性强,可对不同来源檀香的质量进行评价,为其质量控制提供依据。 Objective:To establish the HPLC characteristic chromatogram of methanol extract part of Santalum album,in order to provide the method and basis for evaluating the quality of Santalum album.Methods:The Waters Symmetry C18 column was used,acetonitrile-0.2%phosphoric acid solution was mobile phase with gradient elution,detection wavelength was 205 nm,column temperature was 30℃,the flow rate was 1.0 mL/min;The HPLC characteristic chromatogram was established.Similarity evaluation,cluster analysis and principal component analysis were processed.The content of essential oil in each sample was determined.Results:The characteristic chromatogram of 13 batches of Santalum album samples was established,and eight common peaks were marked.The similarity of each sample was between 0.362 and 0.993 with great difference.The clustering analysis and principal component analysis results were consistent.The volatile oil content of 13 batches of Santalum album samples was between 0.58%and 5.52%,and the volatile oil contents of S3 and S4 samples were relatively low.The result of similarity evaluation was consistent with that of the determination of volatile oil content.Conclusion:The HPLC characteristic chromatogram detection method of Santalum album is established in this experiment,which is simple and rapid with strong specificity,and can better reflect the quality of Santalum album from different sources,and provide the basis for its quality control.
