miR-198在肝细胞癌组织中的表达

Expression of miR-198 in hepatocellular carcinoma

目的探讨肝细胞癌(hepatocellular carcinoma,HCC)组织中miR-198表达及意义。方法HCC患者65例,取手术切除HCC组织及癌旁组织分别为HCC组和癌旁组,采用实时定量PCR法检测2组miR-198mRNA相对表达量;依据癌组织中miR-198mRNA相对表达量将HCC组分为miR-198mRNA高表达组20例,miR-198mRNA低表达组45例,均给予标准方案治疗,比较2组临床病理特征,随访观察2组总生存期。结果HCC组miR-198mRNA相对表达量(1.50±0.82)低于癌旁组(2.11±0.48)(P<0.05);miR-198mRNA低表达组肿瘤中低分化(77.78%)、肿瘤直径≥5cm(71.11%)、有淋巴结转移(75.56%)、有远处转移(80.00%)、TNM分期Ⅲ~Ⅳ期(80.00%)、有门静脉癌栓(68.69%)、有血管浸润(71.11%)、有肿瘤包膜浸润比率(73.33%)高于miR-198mRNA高表达组(25.00%、45.00%、40.00%、50.00%、55.00%、35.00%、40.00%、30.00%)(P<0.05),年龄、性别比例与miR-198mRNA高表达组比较差异无统计学意义(P>0.05);miR-198mRNA低表达组总生存期[(7.63±2.65)个月]较miR-198mRNA高表达组[(20.34±2.73)个月]短(P<0.05)。结论HCC组织miR-198表达明显降低,miR-198低表达可促进肿瘤进展。

Objective To investigate the expression and significance of miR-198in hepatocellular carcinoma(HCC)tissue.Methods The relative expression of miR-198mRNA was detected by real-time quantitative PCR technique in HCC tissue(HCC group)and adjacent tissue(adjacent group)from 65HCC patients.HCC group was redivided into 20 patients with high miR-198mRNA level(high-level group)and 45patients with low miR-198mRNA level(low-level group),and received standard regimen.The clinicopathological characteristics were compared between two groups,and the overall survival was followed up.Results The relative expression of miR-198mRNA was significantly lower in HCC group(1.50±0.82)than that in adjacent group(2.11±0.48)(P<0.05).Low-level group had higher percentages of moderately and poorly differentiated tumor,tumor diameter≥5cm,lymph node metastasis,distant metastasis,TNM stageⅢ-Ⅳ,portal vein tumor thrombus,vascular infiltration and tumor capsule infiltration(77.78%,71.11%,75.56%,80.00%,80.00%,68.69%,71.11%,73.33%)than high-level group(25.00%,45.00%,40.00%,50.00%,55.00%,35.00%,40.00%,30.00%)(P<0.05),and there were no significant differences in the age and sex ratio between two groups(P>0.05).The overall survival was significantly shorter in low-level group((7.63±2.65)months)than that in high-level group((20.34±2.73)months)(P<0.05).Conclusion The expression of miR-198 decreases greatly in HCC tissues,which could promote the progress of HCC.
