摘要
目的采用免疫组织化学法了解FGFR1在我国乳腺癌患者中的表达情况,探讨FGFR1与乳腺癌的发生发展、预后和临床病理学参数的关系,以期找到揭示乳腺癌临床病理特征和预后的可靠分子指标,对乳腺癌预后的预测提供有益的帮助。方法收集潍坊市人民医院2015年6月1日~2017年12月1日收治的有明确病理诊断及分子标记物结果(ER、PR、Her-2)的Ⅰ~Ⅲ期可手术的乳腺癌病例共67例,并选择正常乳腺组织30例作对照。采用免疫组化S-P法进行FGFR1测定。对所有乳腺癌病例进行随访,最后根据随访资料分析FGFR1与乳腺癌临床病理特征的关系。用SPSS13.0统计学软件进行统计分析,计数资料采用χ^2检验,等级资料采用非参数检验,两变量之间的关系采用Spearman等级相关分析。结果乳腺癌组织中FGFR1显著高于正常乳腺组织(P<0.05)。FGFR1的表达量在Ⅰ期和Ⅱ期、Ⅰ期和Ⅲ期、Ⅱ期和Ⅲ期间差异均有统计学意义(P<0.05)。FGFR1在淋巴结转移阳性患者中的表达明显高于淋巴结阴性患者,差异有统计学意义(P<0.05)。结论 FGFR1的表达率与乳腺癌病理类型、组织学分级及淋巴结转移均有关,而与肿瘤大小、年龄无关。FGFR1表达率在淋巴结阳性者高于阴性者。FGFR1的高表达可能与乳腺癌的恶性生物学行为密切相关,可能提示预后不良。
Objective In this study,using immunohistochemical staining for the expression of FGFR1 in invasive ductal carcinoma of breast in China,and explore the relationship between FGFR1,survivin and clinicopathological parameters,in order to guide the treatment of breast cancer,and to predict the prognosis of breast cancer.Methods In this study,collected cases which diagnosised invasive ductal carcinoma of the breast by the Weifang People’s Hospital pathologist,have results of routine pathological parameters(ER,PR,Her-2) and complete clinical data between June 1,2015 to December 1,2017,and got 67 cases of surgical patients’ data.Using the SP immunohistochemical techniques(ICH) detection the expression of FGFR1 protein.According the information available,analysis the relationship between FGFR1 and clinicopathological parameters,clinical pathological features,and analysis of the correlation between the two.Upload the follow-up data accurately to the SPSS13.0 statistical package to do the statistical analysis,the enumeration data was analyzed by χ^2 test or Fisher’s exact test,the ranked data was by non-parametric test,the relationship between the two variables using the Spearman rank correlation.Clinical data with the percentage or median or average for the statistical description.Results The expression of FGFR1 in breast cancer tissue was significantly higher than that in normal breast tissue(P<0.05).There were significant differences in the expression of FGFR1 between stage Ⅰ and Ⅱ,stage Ⅰ and Ⅲ,stage Ⅱ and Ⅲ(P<0.05).The expression of FGFR1 in patients with positive lymph node metastasis was significantly higher than that in patients with negative lymph node metastasis(P<0.05).Conclusion The expression rate of FGFR1 was correlated with pathological type,histological grade and lymph node metastasis of breast cancer,but not with tumor size and age.The expression rate of FGFR1 was higher in lymph node-positive patients than in negative patients.The high expression of FGFR1 may be closely related to the malignant biological behavior of breast cancer,which may indicate poor prognosis.
