摘要
目的分析甲状腺微小乳头状癌(PTMC)中长链非编码RNA(lncRNA)MALAT1和成纤维细胞生长因子2(FGF2)的表达及其与PTMC临床病理特征的关系。方法收集2014年9月至2018年2月行手术治疗的63例PTMC患者的癌组织和癌旁正常组织。采用实时荧光定量PCR(QPCR)检测MALAT1和FGF2的表达水平,分析MALAT1和FGF2的相关性,分析MALAT1和FGF2表达与PTMC临床病理特征的关系。结果PTMC组织中MALAT1表达水平为3.54±0.10,高于癌旁正常组织的1.04±0.05;FGF2表达水平为2.78±0.10,高于癌旁正常组织的0.97±0.05,差异具有统计学意义(P<0.05)。PTMC组织中MALAT1和FGF2表达呈正相关(r=0.738,P=0.001)。MALAT1表达与肿瘤大小及包膜侵犯有关(P<0.05),FGF2表达与肿瘤大小、病灶数、双侧病变、中央淋巴结转移、包膜侵犯及合并NT有关(P<0.05)。结论PTMC中MALAT1和FGF2呈高表达,与PTMC增殖、转移和侵袭密切相关。
Objective To analyze the expression of long-chain non-coding RNA(lncRNA)MALAT1 and fibroblast growth factor 2(FGF2)in thyroid micropapillary carcinoma(PTMC)and their relationship with the clinicopathological characteristics of PTMC.Methods Cancer tissues and adjacent normal tissues of 63 patients with PTMC who underwent surgical treatment from September 2014 to February 2018 were collected.Real-time fluorescence quantitative PCR(QPCR)was used to detect the expression of MALAT1 and FGF2.The correlation between the expression of MALAT1 and FGF2 was analyzed.The relationship between the expression of MALAT1 and FGF2 and the clinicopathological features of PTMC was analyzed.Results The expression level of MALAT1 in PTMC tissues was 3.54±0.10,which was higher than 1.04±0.05 in normal tissues adjacent to cancer;the expression level of FGF2 in PTMC tissues was 2.78±0.10,which was higher than 0.97±0.05 in normal tissues adjacent to cancer.The difference was statistically significant(P<0.05).The expression of MALAT1 and FGF2 in PTMC was positively correlated(r=0.738,P=0.001).The expression of MALAT1 was correlated with tumor size and capsular invasion(P<0.05);and the expression of FGF2 was correlated with tumor size,number of lesions,bilateral lesions,central lymph node metastasis,capsular invasion and NT(P<0.05).Conclusion High expression of MALAT1 and FGF2 in PTMC is closely related to proliferation,metastasis and invasion of tumors.
