摘要
目的建立测定罗汉松种子挥发油中川芎嗪含量的高效液相色谱法。方法色谱柱为GL Inertsil ODS-3 C18柱(250 mm×4.6 mm,5.0μm),流动相为甲醇-0.1%磷酸溶液(60∶40,V/V),流速为0.8 m L/min,检测波长为298 nm,柱温为室温。结果川芎嗪进样质量浓度在0.38~22.72μg/mL范围内与峰面积线性关系良好(r=0.999 8);平均回收率为99.42%,RSD为1.31%(n=6)。结论该方法简便、准确、重复性好,可用于测定罗汉松种子挥发油中川芎嗪的含量。
Objective To establish an HPLC method for the content determination of ligustrazine in volatile oil from the seed of Podocarpus macrophyllus.Methods The chromatographic column was GL Inertsil ODS-3 C18 column(250 mm×4.6 mm,5.0μm),the mobile phase was methanol-0.1%phosphoric acid solution(60∶40,V/V),the flow rate was 0.8 mL/min,the detection wavelength was 298 nm and the column temperature was room temperature.Results The ligustrazine showed good linear relationship with peak area in the range of 0.38-22.72μg/mL(r=0.999 8),the average recovery was 99.42%and RSD was 1.31%(n=6).Conclusion The method is simple,accurate and repeatable,which can be used for the content determination of ligustrazine in volatile oil from the seed of Podocarpus macrophyllus.
