RANK基因甲基化与新疆汉、维吾尔、哈萨克族老年男性肥胖相关性研究

Correlation between RANK Gene DNA Methylation and Obesity in Male Elderly Uygur,Kazak and Han Nationality in Xinjiang

摘要 目的分析RANK基因甲基化变异与肥胖的相关性。方法资料来源于2018年4~5月对新疆维吾尔自治区木垒县、洛浦县65岁以上的汉族、维吾尔族、哈萨克族居民流行病学调查研究数据,随机抽取32例汉族、28例维吾尔族、30例哈萨克族老年男性,各分为病例组和对照组。流行病学调查采用问卷调查、体格检查和实验室检测获取研究对象的一般资料及血生化指标。应用EpiTect Fast Bisulfite Conversion Kits进行基因组甲基化修饰,采用BiQ Analyzer分析软件对RANK基因组标准序列和测序序列进行比对。采用协方差分析,在校正年龄、吸烟、饮酒、高血压发生率及血脂后,分析对照组、病例组RANK基因CpG岛甲基化率差异。结果RANK基因CpG岛甲基化率在3个民族间比较差异无统计学意义;在校正年龄、吸烟、饮酒、高血压发生率及血脂后,在新疆维吾尔自治区汉族、维吾尔族、哈萨克族老年男性中,对照组RANK基因CpG岛甲基化率高于病例组,差异有统计学意义(P<0.05)。结论在新疆汉族、维吾尔族、哈萨克族老年男性中,RANK基因甲基化率降低可能与肥胖相关。 Objective To investigate the correlation between RANK gene methylation and obesity by case-control study.Methods Data were collected from the epidemiological survey data of Han,Uygur,Kazak residents aged 65 and over in LuoPu county and MuLei county,Xinjiang from April 2018 to May 2018,which 32 Han,28 Uygur and 30 Kazakh elderly men were randomly selected and divided into case group and control group.General information and blood biochemical indexes of the subjects were obtained by questionnaire survey and laboratory test.The genomic methylation modification was performed on EpiTect Fast Bisulfite Conversion Kits,BiQ Analyzer was used to compare the RANK genome standard sequence and sequencing sequence.The methylation rate of RANK gene CpG island in the control group was analyzed by covariance analysis after adjusting the prevalence rate of age,smoking,drinking and hypertension and blood lipid.Results Among elderly Han,Uygur and Kazak nationality males of Xinjiang.The methylation rate of RANK gene CpG island had no statistics significance(P>0.05).After adjusting the prevalence rate of age,smoking,drinking and hypertension and blood lipid among elderly Han,Uygur and Kazak nationality males of Xinjiang.The methylation rate of RANK gene CpG island in the control group was higher than that in the case group(P<0.05).Conclusion Among elderly Han,Uygur and Kazak nationality males of Xinjiang,a slight decrease in RANK methylation may be associated with obesity.
