马比木根茎提取物黄酮和三萜含量及其降糖活性 被引量:2

Total Tlavonoid and Triterpene Contents of Extracts From Root and Stem of NothapodytesPittosporoides

摘要 为探索马比木根茎中总黄酮和总三萜含量及其胰岛素抵抗降糖活性,对其采用甲醇提取测定根、茎总黄酮和总三萜含量,用HepG2细胞制备胰岛素抵抗降糖模型,进行了降糖活性的检测。结果显示:根中总黄酮含量(204.75 mg/g)高于茎中含量(114 mg/g);总三萜的含量根中(145 mg/g)低于茎中含量(225.17 mg/g)。采用MTT法对HepG2细胞进行活力检测,排除提取物溶剂及提取物的细胞毒性,且细胞活性良好,具有消耗葡萄糖的能力。与阳性药二甲双胍进行降糖活性对照,根-茎提取物的不同剂量的降糖活性均优于阳性药。根提取物的降糖活性优于茎提取物,这与总黄酮的含量结果一致,说明降糖活性与总黄酮具有正相关的作用。实验结果说明了总黄酮的降糖活性优于总三萜的降糖活性。这为马比木资源的再利用提供新的思路和途径,既开拓了马比木功效利用的短板,又减少了马比木资源的过度消耗和浪费。 In order to explore total flavonoids and triterpenes contents of extracts from root and stem of Nothapodytes Pittosporoides,and their insulin resistance hypoglycemic activity,the contents of total flavonoids and triterpenes from root and stem of Nothapodytes Pittosporoides were determined by methanol extraction,and the insulin resistance hypoglycemic model was prepared by HepG2 cells to detect the hypoglycemic activity.The results showed that the total flavonoids content(204.75mg/g)in roots of Nothapodytes Pittosporoides was higher than that(114mg/g)in stems of NothapodytesPittosporoides.The total triterpene content(145mg/g)was lower in roots of Nothapodytes Pittosporoides,than(225.17mg/g)in stems of Nothapodytes Pittosporoides.HepG2 cells were tested by MTT method to eliminate the cytotoxicity of the solvent and extract,and the cell activity was good that showing the ability to consume glucose.Compared with the positive drug metformin,the hypoglycemic activity of different doses of root-stem extract of Nothapodytes Pittosporoides were better than that of the positive drug.The hypoglycemic activity of root extract of Nothapodytes Pittosporoides was better than that of stem extract of Nothapodytes Pittosporoides,which was consistent with the content of total flavonoids.It indicated that the hypoglycemic activity had a positive correlation with total flavonoids.The results showed that the hypoglycemic activity of total flavonoids was better than that of total triterpenes.This paper provided a new idea and way for the reuse of Nothapodytes Pittosporoidesresources,which not only opened the shortcomings of Nothapodytes Pittosporoides efficiency utilization,but also reduced the excessive consumption and waste of Nothapodytes Pittosporoides!
