摘要 为测定金针菇中总的粗多糖含量,采用热水提取法提取了金针菇多糖,通过二硝基水杨酸显色法,以葡萄糖为对照,测定了还原糖和总糖的含量,二者差值即为总的粗多糖含量。结果表明:金针菇中粗多糖含量为0.05 mg/mL;DNS法的重复性、稳定性、加样回收均较好,提高了试验的准确度和效率。DNS法操作简单,所受干扰较少,适合于快速测定金针菇样品中总的粗多糖含量。 [Objective]To determine thecontentof total polysaccharide in flammulinavelutiper.[Method]Flammulinavelutiper polysaccharide was extracted by hot water extraction.The content of reducing sugar and total sugar was determined by dinitrosalicylic acid colorimetry and glucose as control.The difference between the two was the total crude polysaccharide content.[Result]The contentof totalpolysaccharide inflammulinavelutiper was 0.05mg/mL.The DNS method has good repeatability,stability,and sample recovery,which improves the accuracy and efficiency of the test.[Conclusion]The DNS method,with simple procedure and little interference,was suitable for rapid determination the content of total polysaccharide in flammulinavelutiper.
