摘要 为建立紫叶稠李组织培养快速繁殖方法,对紫叶稠李重要种质材料的保存扩繁提供技术支持。本研究以紫叶稠李为材料,以MS培养基为基本培养基,通过不同种类和浓度的植物生长调节剂配比试验,筛选紫叶稠李快速繁殖与生根的最佳培养基组成。结果表明:紫叶稠李试管苗最佳增殖培养基为MS+2~3 mg·L^-1 6-BA+0.1 mg·L^-1 NAA+30 g·L^-1蔗糖+8 g·L^-1琼脂,最佳生根培养基为1/2 MS+0.1 mg·L^-1 IBA+30 g·L^-1蔗糖+8 g·L^-1琼脂,生根诱导20 d,生根率达90%。将生根的试管苗在室外进行炼苗后,移栽到营养钵中,成活率达到90%以上。 In order to establish a rapid propagation method for tissue culture of Padus virginiana,technical support was provided for the preservation and propagation of important germplasm materials of Padus virginiana.In this study,Padus virginiana was used as material,MS medium was used as basic medium,and the optimum medium composition for rapid propagation and rooting of Padus virginiana was screened through the ratio test of plant growth regulators of different kinds and concentrations.The results showed that the optimum medium for multiplication was MS+2-3 mg·L^-1 6-BA+0.1 mg·L^-1 NAA+30 g·L^-1 sucrose+8 g·L^-1 agar,and the optimum medium for rooting was 1/2 MS+0.1 mg·L^-1 IBA+30 g·L^-1 sucrose+8 g·L^-1 agar.Rooting induction lasted for 20 days and the rooting rate reached 90%.After the rooting test-tube seedlings were refined outdoors,they were transplanted into nutrient bowls,and the survival rate reached more than 90%.
