摘要
目的:探讨芪防膝痹颗粒对大鼠膝骨关节炎(KOA)疼痛的影响。方法:选取16周龄SD雌性大鼠40只,分为对照组、模型组、塞来昔布和芪防膝痹颗粒组,每组10只。除对照组外其余行前交叉韧带切除建立KOA,术后分别给予0.9%氯化钠溶液、塞来昔布和芪防膝痹颗粒灌胃,8周后取材。Electric Von Frey疼痛仪于术前及术后每周测1次疼痛阈值,Micro-CT扫描后行HE、番红-快绿染色观察滑膜及软骨的改变,免疫组织化学观察IL-1β、TNF-α、VEGF的表达,ELISA测血清IL-6水平。结果:与模型组比较,芪防膝痹颗粒组KOA疼痛阈值显著升高(P<0.05),滑膜炎症和软骨破坏也明显减轻,VEGF在软骨边缘、滑膜以及IL-1β、TNF-α在软骨中表达均降低(P<0.05)。结论:芪防膝痹颗粒可以减轻KOA疼痛和延缓软骨退变,可能与减少滑膜中VEGF表达有关。
Objective: To investigate the effect of Qifang Xibi Granules on pain of knee osteoarthritis(KOA) in rats. Methods: A total of 40 sixteen weeks of SD female rats were randomly divided into the control group, model group, celecoxib group and Qifang Xibi Granules group with 10 rats in every group. KOA was established by anterior cruciate ligament excision except the control group. Saline, celecoxib and Qifang Xibi Granules were given to the stomach after the operation. Samples were taken after 8 weeks. Electric Von Frey pain meter was used to measure pain before and after operation once a week. After micro-CT scanning, the changes of synovium and cartilage were observed by HE and SO. The expressions of IL-1β TNF-α and VEGF were observed by immunohistochemistry. Serum IL-6 levels were measured by ELISA. Results: Compared with the model group, the pain threshold of KOA in Qifang Xibi Granules group was significantly increased(P<0.05), synovitis and cartilage destruction in knee joints were significantly reduced, and the expression of VEGF in cartilage edge, synovium, IL-1β and TNF-α were all decreased(P<0.05). Conclusion: Qifangxibi Granule can relieve KOA pain and delay cartilage degeneration, which may be related to the decrease of the expression of vascular endothelial growth factor in synovium.
