黄连解毒汤对上火牙龈炎大鼠β-防御素的影响 被引量:5

Influences of Huanglian Jiedu Decoction on the expression of β-defensins in rat gingivitis

摘要 目的:构建大鼠感染致上火牙龈炎模型,观察黄连解毒汤对牙龈组织β-防御素1(BD-1)和β-防御素2(BD-2)表达的影响。方法:采用牙龈黏膜下注射脂多糖法构建大鼠牙龈炎模型,模拟中医上火证候,大鼠分为6组:假手术组,模型组,黄连解毒汤低、中、高剂量组,阳性对照药组,每组10只。各组予相应药物干预。实验结束后,刮取牙龈组织,qRT-PCR法和Western blotting法分别在基因和蛋白水平检测用药前后BD-1和BD-2的表达。结果:与假手术组比较,模型组牙龈组织中BD-1和BD-2 mRNA及蛋白表达均显著降低(P<0.01);与模型组比较,黄连解毒汤低、中、高剂量组BD-1和BD-2 mRNA及蛋白表达均显著升高(P<0.05,P<0.01)。结论:β-防御素与上火牙龈炎密切相关,BD-1和BD-2可能是黄连解毒汤潜在的火热证候治疗靶点。 Objective: To construct rat shanghuo gingivitis model and observe the effects of Huanglian Jiedu Decoction(HLJD) on expression of β-defensin-1(BD-1) and β-defensin-2(BD-2) in gingival tissues. Methods: Rat shanghuo gingivitis model was established by LPS injection. Adult male rats were divided into six equal groups with 10: sham group;model group;HLID low-dose, middle-dose, and high-dose group, positive control group. Each group was given corresponding drug intervention. After experement, the rats were sacrificed and gingival tissue samples were randomly determined by qRT-PCR and Western blotting method. Results: Compared with sham group, both genetic and proteic expression of BD-1 and BD-2 were obviously decreased in model group(P<0.01). Compared with model group, HLJD low, middle and high group could increase mRNA and protein expression level of HLJD significantly(P<0.05, P<0.01). Conclusion: β-defensins are closely related with shanghuo gingivitis, and BD-1 and BD-2 may be identified as potential therapeutic targets.
