摘要
目的:探讨芍甘附子汤加味对人类风湿关节炎成纤维样滑膜细胞(HFLS-RA)HIF-VEGF-ANG轴的影响,为芍甘附子汤加味治疗类风湿关节炎的临床应用提供实验基础。方法:将HFLS-RA细胞进行传代培养,选用第4~8代HFLS-RA进行研究。实验分为空白组、诱导组、对照组和芍甘附子汤加味大、中、小剂量组。各组予相应药物干预。MTT法检测HFLS-RA细胞的增殖,qPCR法测定HFLS-RA细胞培养上清液中HIF-VEGF-ANG的表达。结果:芍甘附子汤加味可抑制HFLS-RA增殖,降低细胞培养上清液中HIF-1α、VEGF、VEGF2、Ang-1、Ang-2的表达。结论:芍甘附子汤加味通过抑制HFLS-RA增殖及调控HIF-VEGF-ANG的表达发挥作用。
Objective: To explore the effect of modified Shaogan Fuzi Decoction on the HIF-VEGF-ANG axis of rheumatoid arthritis fibroblast-like synovial cell(HFLS-RA), and provide experimental basis for the clinical application of Shaogan Fuzi Decoction in the treatment of RA. Methods: The purchased HFLS-RA cells were subcultured and the 4 th to 8 th generation HFLS-RA was used for study. The experiments was divided into blank group, induction group, control group, and modified Shaogan Fuzi Decoction large, medium and small dose treatment groups. After intervention, the morphology of cell growth was observed under electron microscope, the proliferation of HFLS-RA cells were detected by MTT, and the expression of HIF-VEGF-ANG in the culture supernatant of HFLS-RA cells was determined by qPCR. Results: Modified Shaogan Fuzi Decoction can inhibit the proliferation of HFLS-RA and reduce the expression of HIF-1α, VEGF, VEGF2, Ang-1 and Ang-2 in the cell culture supernatant. Conclusion: Modified Shaogan Fuzi Decoction can play a role in inhibiting the proliferation of HFLSRA and regulating the expression of HIF-VEGF-ANG.
