摘要
目的:探讨薏苡仁不同组分对脾虚湿阻证大鼠细胞因子-细胞因子受体相互作用的影响及关键靶点。方法:以薏苡仁不同组分作用于脾虚湿阻证大鼠,通过其肠全基因表达谱测定,对筛选的差异基因后进行KEGG分析,并对富集到的细胞因子-细胞因子受体相互作用通路基因进行主成分分析和Hierarchical Clustering聚类分析。结果:模型大鼠与空白大鼠注释到细胞因子-细胞因子受体相互作用的差异基因为29条;对差异基因主成分分析的Loading和Score进行相关性分析显示,蛋白组和淀粉组治疗作用较弱而油组和多糖组治疗效果较好;效果最为显著的前10位是Tnfrsf9、Cd27、Ccl22、Cd40、Ccl20、Il21r、Ccr7、Tnfsf9、Cxcl13、Cxcr5;Hierarchical Clustering聚类分析显示,第Ⅰ、Ⅲ大类的差异基因发挥了主要作用;KEGG分析显示,细胞因子-细胞因子受体相互作用差异基因主要涉及的信号通路出现在趋化因子家族和TNF家族中。结论:脾虚湿阻证大鼠细胞因子-细胞因子受体相互作用紊乱,薏苡仁各个组分,尤其是薏苡仁油能够很好的调理混乱的细胞因子体系,尤其是趋化因子,改善机体炎性反应及免疫功能,这可能是薏苡仁健脾利湿的机制所在。
Objective: To explore the effects and key targets of different coix seed components on the cytokines-cytokine receptors interaction(CCI) in syndrome of damp retention due to spleen deficiency(SDSD). Methods: The SDSD rats model were treated by different coix seed components to detect intestinal gene expression profile by gene chip. The different express genes(DEGs) of the intestinal were enrichment by Kyoto encyclopedia of genes and genomes(KEGG). Then the related DEGs of CCI were analyzed by PCA analysis and Hierarchical Clustering analysis. Results: There were 29 DEGs in CCI between the SDSD rat and control rat. The protein group and starch group showed weaker treatment effect while the oil group and the polysaccharide group showed better effect through the correlation analysis of loading and score of the PCA of the DEGs. The top 10 DEGs were Tnfrsf9, Cd27, Ccl22, Cd40, Ccl20, Il21 r, Ccr7, Tnfsf9, Cxcl13, Cxcr5. TheⅠ, Ⅲ category DEGs played the major role through hierarchical cluster analysis. KEGG analysis showed that the main signaling pathways involved in CCI were in the chemokine family and tumor necrosis factor family. Conclusion: The CCI appears imbalance in the SDSD rats. The coix seed components, especially oil can regulate the CCI chaos, especially chemokine, improve the body’s inflammatory reaction and immune function. It may be the mechanisms of coix seed to treat SDSD.
