沙棘宫愈方有效部位对人宫颈癌Hela细胞增殖的影响及其配伍关系研究

Study on the effects of effective fraction of Shaji Gongyu Formula on proliferation of human cervical cancer Hela cells and its compatibility relationship

摘要 目的:考察对沙棘宫愈方三种有效部位抗人宫颈癌Hela细胞增殖的影响及其配伍关系。方法:采用体外MTT法测定了沙棘籽油、苦参碱、蛇床子素这3种有效部位对人宫颈癌Hela细胞增殖的抑制作用;采用基线等比增减设计法探讨苦参碱与蛇床子素二者配伍的最佳比例。结果:苦参碱、蛇床子素提取物能显著抑制人宫颈癌Hela细胞的增殖(P<0.01,P<0.05)。不同配比的苦参碱和蛇床子素提取物对人宫颈癌Hela细胞有显著的抑制作用(P<0.01),其中苦参碱1.802mg/mL与蛇床子素提取物0.035mg/mL配伍后对Hela细胞的抑制作用最强。结论:苦参碱与蛇床子素提取物均具有明显的抗人宫颈癌Hela细胞增殖的作用,而沙棘籽油没有。苦参碱1.802mg/mL与蛇床子素提取物0.035mg/mL配伍后抗Hela的效果最好。 Objective: To investigate the effects of three effective fractions of Shaji Gongyu Formula on the proliferation of Hela cells and the compatibility of the three effective fractions. Methods: The inhibitory effect of Seabuckthorn seed oil, matrine and osthole extracte extract to Hela was detected by MTT assay in vivo;The base line equivalent design method was adopted to investigate the best compatibility proportion of matrine and osthole. Results: Both matrine and osthole extract can significantly inhibit the proliferation of Hela(P<0.01, P<0.05). We find that different ratios of matrine and osthole extract can significantly affect the suppression of Hela(P<0.01), the best combination concentration of which are 1.802 mg/mL of matrine and 0.035 mg/mL of osthole extract. Conclusion: Both matrine and osthole extract inhibit the proliferation of Hela, while Seabuckthorn seed oil cannot inhibit;the combination of matrine 1.802 mg/mL and osthole extract extract 0.035 mg/mL show the best inhibitory effect.
