芍苓汤改善豚鼠银屑病皮损活性成分初探 被引量:2

Preliminary study on active ingredients of Shaoling Decoction which improved propranolol-induced psoriasis-like lesion in guinea pigs

摘要 目的:探究芍苓汤改善豚鼠银屑病皮损的药效作用物质基础。方法:将芍苓汤复方按照极性分为乙酸乙酯部位(Ⅰ)、水洗脱部位(Ⅱ)、50%乙醇部位(Ⅲ)、95%醇洗脱部位(Ⅳ)考察其对盐酸普萘洛尔诱导豚鼠银屑病样模型的改善作用,以Baker评分量化药效结果;对芍苓汤不同部位成分进行分析,考察药效-成分相关性。结果:与正常组比较,模型组Baker评分显著增加(P<0.01)。芍苓汤和甲氨蝶呤均能改善豚鼠耳部病变,Baker评分显著降低(P<0.01),Baker评分抑制率分别为47.7%、42.2%、60.0%和61.3%。成分分析结果显示,部位Ⅰ主要含有落新妇苷、芍药苷及其异构体,部位Ⅲ主要含芍药苷。部位Ⅱ和Ⅳ在当前色谱条件下无可检测成分。结论:芍苓汤的主要活性成分为芍药苷、落新妇苷及其异构体,水洗脱部位具活性,其成分有待进一步研究。 Objective: To explore the material basis of Shaoling Decoction(SLD) for antipsoriatic efficacy. Methods: SLD was divided into ethyl acetate part-(Ⅰ), water eluent-(Ⅱ), and 50% ethanol eluent-(Ⅲ), 95% alcohol eluent-(Ⅳ) according to their polarities. The Baker-score was used to quantify the effect of different parts on improving the propranolol-induced psoriasislike model of guinea pigs, and the constituents of different parts were analyzed, then the efficacy-components correlations were investigated. Results: Compared with the normal group, the Baker-score of the model group was significantly increased(P<0.01). SLD and Methotrexate all improved guinea pigs’ ear lesion, and the Baker scores was decreased by 47.7%, 42.2%, 60.0% and 61.3%, respectively(P<0.01). The results of constituent analysis showed that Part Ⅰ mainly contained astilbin, paeoniflorin and their isomers and part Ⅲ mainly contained paeoniflorin. Part Ⅱ and Ⅳ had no detectable components under current chromatographic conditions. Conclusion: The main bioactive constituents of the SLD were paeoniflorin, astilbin and their isomers. The active constituents of water elution need to be further explored.
