摘要
目的探讨丹参酮ⅡA(TanⅡA)对肝细胞癌的保护作用,并阐明其可能的机制。方法分别用5μg/ml、10μg/ml TanⅡA处理肝癌细胞HepG2 24、48 h后,采用噻唑兰法、克隆实验、流式细胞术、划痕实验与Transwell实验检测细胞增殖、周期、侵袭和转移情况,采用Western blot法检测相关通路蛋白表达。结果与对照组比较,TanⅡA对肝癌细胞HepG2细胞的增殖具有明显的抑制作用,随浓度增加抑制作用逐渐增强,差异有统计学意义(P<0.05);与对照组比较,TanⅡA显著抑制肝癌细胞HepG2的侵袭和迁移,差异有统计学意义(P<0.05);与对照组比较,TanⅡA显著抑制p-Akt表达,降低p-Akt/Akt比率,促进PTEN蛋白表达,差异有统计学意义(P<0.05)。结论TanⅡA抑制肝癌细胞增殖、周期停滞与迁移,其调控机制可能与Akt/PTEN通路有关。
Objective To investigate the protective effect of tanshinoneⅡA(TanⅡA)on hepatocellular carcinoma(HCC),and elucidate its possible mechanism.Methods With 5μg/ml,10μg/ml TanⅡtreatment A HCC cell line HepG2 24 and 48 hours after using thiazole blue,determined by MTT method,the cloning experiment,flow cytometry,scratches and Transwell experiment test cell proliferation,cycle,invasion and metastasis;Western blot was used to detect the expression of related pathway proteins.Results Compared with the control group,TanⅡA HepG2 cell proliferation inhibition,with the increase of concentration of inhibition enhanced gradually,and the difference was statistically significant(P<0.05).Compared with the control group,TanⅡA significant inhibition of HepG2 cell invasion and migration(P<0.05).Compared with the control group,TanⅡA significant inhibition of p-Akt expression,reduce the ratio of p-Akt/Akt and promote the PTEN protein expression(P<0.05).Conclusion TanⅡA inhibits the proliferation,cell cycle arrest and migration of HCC cells,and its regulatory mechanism may be related to Akt/PTEN signal pathway.
