摘要
目的探讨广金钱草总黄酮片对大鼠肾结石的作用及机制。方法采用乙二醇加氯化铵法复制大鼠肾结石模型,观察各组大鼠血清Ca^2+、Mg^2+、肌酐、尿素氮、尿Ca^2+、Mg^2+及肾组织中草酸含量。病理观察肾组织草酸钙结晶进行结晶量分级评分。检测肾组织超氧化物岐化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平。结果与模型对照组比较,广金钱草总黄酮片中、高剂量组血清Ca^2+、肌酐、尿Ca^2+、肾组织草酸、MDA含量、结晶评分显著降低(P<0.05,P<0.01),广金钱草总黄酮片高剂量组血清尿素氮含量显著降低(P<0.05),血清Mg^2+、尿Mg^2+、肾组织SOD含量增高(P<0.05,P<0.01)。结论广金钱草总黄酮对大鼠肾结石具有明显的抑制作用,其机制可能与降低草酸水平、减少尿Ca^2+排泄、减轻脂质过氧化损伤有关。
Objective To explore the effect and the potential mechanism of the total flavonoid tablets of desmodium styracifolium(TFDS)on renal calculus in rats.Methods The model of renal calculus in rats was established by administrating ethylene glycol and ammonia chloride.The content of Ca^2+,Mg^2+,creatinine,urea nitrogen in serum and Ca^2+,Mg^2+in urine were detected as well as oxalic acid in renal tissue.The calcium oxalate crystal was observed pathologically and the grade of crystal deposition was scored.The levels of superoxide dismutase(SOD)and malonaldehyde(MDA)in kidney tissues were also determined.Results Compared with the model control group,in the high-dose TFDS group,the levels of Ca^2+,Mg^2+,creatinine,urea nitrogen in serum and Ca^2+in urine,oxalic acid in renal tissue and MDA,as well as crystal deposition score were all reduced significantly(P<0.05,P<0.01).In the high-dose TFDS group,urea nitrogen in serum was reduced significantly(P<0.05),the serum Ca^2+,urine Mg^2+and SOD in renal tissue were increased(P<0.05,P<0.01).Conclusion TFDS displays the obvious inhibitory effect on renal calculus in the rats,which may be related to the decrease of oxalic acid,the excretion of urine Ca^2+and the alleviation of the injury of lipid peroxidation.
