摘要
目的探讨采用BALB/c小鼠建立子宫内膜异位症模型的可行性及异位病灶纤维化的发生情况。方法选用BALB/c小鼠40只,随机取10只作为空白组、20只作为实验组,余10只作为供体小鼠。实验组小鼠采用腹腔注射含供体小鼠子宫碎片的生理盐水法建立子宫内膜异位症模型,于造模后第14天和第21天时观察异位病灶生长情况,并取异位病灶进行HE染色、Masson染色观察。结果造模后第14天和第21天时,小鼠子宫内膜异位症模型建立成功率分别为80%(16/20)、90%(18/20);大体观察异位病灶多位于小鼠左盆腹腔,呈单个或多房囊样;HE染色可见异位病灶中有子宫内膜上皮细胞、腺体;Masson染色可见异位病灶有大量纤维沉积生成。结论采用BALB/c小鼠腹腔注射法可成功建立子宫内膜异位症动物模型,且操作简便、成模率高;异位病灶有纤维化生成,可作为子宫内膜异位症纤维化机制和药物疗效研究的可靠动物模型。
Objective It is to investigate the feasibility of establishing endometriosis(EMs)model and the occurrence of ectopic lesion fibrosis in BALB/c mice.Methods Forty BALB/c mice were selected,in which 10 ones were randomly divided as blank group,20 ones as experimental group and 10 ones as donor mice.In the experimental group,the animal model of EMs was established by intraperitoneal injection of normal saline containing donor uterus fragments.The growth of ectopic lesions were observed on the 14th and 21st day after modeling,and the ectopic lesions were taken for HE staining and Masson staining.Results On the 14th and 21st day after model establishment,the success rate of EMs model mice was 80%(16/20)and 90%(18/20),respectively.The general observation showed that ectopic lesions were mostly located in the left pelvic cavity of mice with single or multiple cystic lesions.HE staining showed that there were endometrial epithelial cells and glands in ectopic lesions;Masson staining showed that a large number of fibrous deposits were formed in ectopic lesions.Conclusion The EMs animal model can be successfully established by intraperitoneal injection of BALB/c mice.It is easy to operate with a high rate of successful model establishing.The ectopic lesions have fibrosis,which can be used as a reliable animal model for the study on EMs fibrosis mechanism and drug efficacy.
