PAPD4基因缺失影响小鼠的运动功能

摘要 目的研究PAPD4基因敲除对小鼠行为和生理状态的影响。方法按性别、基因型将小鼠分为雄性对照、雄性全敲、雌性对照和雌性全敲。开放场、水迷宫、血糖检测、血压检测、T迷宫、糖水偏好和强迫游泳。结果雌性敲除鼠在开放场中的运动速度(P<0.05)和总运动距离(P<0.001)显著少于雌性对照。雌、雄敲除鼠在水迷宫中的运动速度低于同性对照,且雌性小鼠在第6次和第9次训练的运动速度存在显著低于对照(P 6<0.05,P 9<0.05);雄性小鼠在第4次和第9次训练的运动速度存在显著低于对照(P 4<0.01,P 9<0.05)。此外,雄性敲除鼠的基线血糖水平显著低于雄性对照(P<0.05);禁食16 h,其血糖水平显著高于对照(P<0.05),且禁食前后的血糖改变量显著低于对照(P<0.01)。结论PAPD4基因缺失可能影响小鼠的运动功能,并影响雄性小鼠的血糖水平。 Objective To explore the influence of PAPD4 gene deletion on mouse behaviors and physiologies.Methods Mice were divided into four groups by sex and genotype:male control,male knockout,female control and female knockout.Open field test,Morris water maze,blood glucose test,blood pressure test,T maze,sucrose preference test and forced swimming test.Results In open field test,the locomotion velocity(P<0.05)and the total distance(P<0.001)of female KO mouse were significantly slower than female controls.In Morris water maze test,the locomotion velocity of KO mouse was lower than that in normal controls.Specifically,the velocity of female KO mouse was significantly slower than female controls in the sixth and ninth trial(P 6<0.05,P 9<0.05),and the velocity of male KO mouse was also significantly slower than male controls in the fourth and ninth trial(P 4<0.01,P 9<0.05).Otherwise,the blood glucose of male KO mouse was significantly lower than male normal controls at adlibitum state(P<0.05);but after 16 hours food deprivation,the blood glucose of male KO mouse was significantly higher than male controls(P<0.05),and the change of male KO mouse was significantly lower than male controls(P<0.01).Conclusions Deleting of PAPD4 gene affects locomotion ability of mouse,and this gene may function in regulation of blood glucose level in male mouse.
