Clinical Significance of ALDH3B1 Expression in Acute Lymphoblastic Leukemia

目的明确ALDH3B1基因在急性淋巴细胞白血病(ALL)细胞中的表达情况及其临床意义,并探讨其功能。方法收集43例初治ALL患者的临床资料,检测ALDH3B1的表达水平,分析其临床相关性。在ALL细胞中过表达ALDH3B1,观察其对细胞生物学行为的影响。结果ALL患者中ALDH3B1基因表达水平较非造血系统恶性疾病患者低(P=0.000 6)。初诊时外周血白细胞高的患者ALDH3B1基因表达水平相对低(P=0.035 8)。ALDH3B1基因表达水平低的患者总体生存时间更短(P=0.034 1),且治疗达到完全缓解后再复发的患者ALDH3B1基因表达水平明显低于未复发患者(P=0.046 2)。过表达ALDH3B1的ALL细胞系增殖受到抑制。结论ALL患者中ALDH3B1基因表达水平降低,且ALDH3B1基因表达水平低的ALL患者预后不良。ALDH3B1在ALL的发病过程中可能起抑癌作用。

Objective To investigate the expression of ALDH3B1 gene,determine its clinical significance in acute lymphoblastic leukemia(ALL)cell lines,and explore its function.Methods The clinical data of 43 patients with newly diagnosed ALL were collected.The expression level of ALDH3B1 was detected and its correlation with clinical characteristics of patients with ALL was analyzed.ALDH3B1 was overexpressed in ALL cells,and its effect on cell biological behavior was observed.Results The expression level of ALDH3B1 gene in patients with ALL was lower than that in normal controls(P=0.000 6).The expression level of ALDH3B1 gene was relatively lower in patients with high peripheral blood leukocyte counts at the time of initial diagnosis(P=0.035 8).Patients with lower ALDH3B1 gene expression had shorter overall survival(P=0.034 1),and ALDH3B1 gene expression was significantly lower in patients who had relapse after complete remission(P=0.046 2).The proliferation of ALL cell lines overexpressing ALDH3B1 was inhibited.Conclusion The expression level of ALDH3B1 gene is decreased in patients with ALL,and patients with ALL with low ALDH3B1 gene expression have poor prognosis.ALDH3B1 may play a role in tumor suppression during the pathogenesis of ALL.
