摘要
为进一步研究基于标记的唇[鱼骨]Hemibarbus labeo苗种放流效果评估工作,以安谷水电站鱼类增殖放流站实际工作中的两个唇[鱼骨]家系为研究对象,采用6个多态性微卫星标记对两个唇[鱼骨]家系开展了亲子关系鉴定分析。结果显示:6个微卫星标记共检测出37个等位基因,观测杂合度和期望杂合度分别为0.361~0.952(平均值为0.776)和0.302~0.795(平均值为0.589);多态信息含量为0.262~0.767(平均值为0.526);Cervus软件分析显示,在双亲均未知的情况下,唇[鱼骨]6个微卫星位点的累计非亲权排除率(CEP)为99.09%;两个家系中的72个子代个体,正确分配到对应家系父母本(12个亲本)的总体比例为95.83%。研究表明,本研究中采用微卫星标记方法来开展唇[鱼骨]家系亲子鉴定的分析,可为唇[鱼骨]的增殖放流效果评估提供技术支撑。
In the present study,six polymorphic microsatellite markers were first used in parentage analysis for the two families of Hemibarbus labeo at Angu Proliferation Releasing Station.A total of 37 alleles were obtained,with the observed heterozygosis ranging from 0.361 to 0.952(mean value 0.776),expected heterozygosis varying from 0.302 to 0.795(mean value 0.589),and polymorphic information content(PIC)ranging from 0.262 to 0.767(mean value 0.526).The Cervus analysis indicated that there was combined non-exclusion probability(CEP)of 99.09%if parents were both unknown.The parentage allocation rate was 95.83%(12 parents)in 72 offsprings of the two families,with above 95%of both CEP and parentage allocation rate,which is a basic requirement value for parentage analysis.The finding shows that these six microsatellite markers are useful for parentage identification,and provides technical support for evaluating stock enhancement in Hemibarbus labeo in the future.
