桃胶多糖及其协同EGCG抗醉解酒作用研究 被引量:5

Study on the Effect of Peach Gum Polysaccharides in Combination with Epigallocatechin Gallate on Anti-drunk and Anti-alcohol Effect

摘要 通过观察桃胶多糖(peach gum polysaccharide,PGP)对急性乙醇性胃粘膜损伤的保护作用,探讨桃胶多糖协同表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)对小鼠的抗醉解酒作用及其生物学机制。体外试验以透析袋模拟胃膜,研究不同浓度PGP抑制乙醇扩散的影响。动物实验以小鼠为对象,给予60%食用乙醇灌胃造模,不同浓度的EGCG与PGP协同干预后,观察小鼠的醉酒耐受时间、醒酒时间变化,评估小鼠胃粘膜损伤情况,测定小鼠血清乙醇浓度变化,测定小鼠胃组织氧化应激相关分子含量变化,测定小鼠肝组织乙醇脱氢酶(alcohol dehydrogenase,ADH)、乙醛脱氢酶(acetaldehyde dehydrogenase,ALDH)含量变化。体外及体内试验结果表明PGP可显著抑制乙醇扩散,有效延长小鼠醉酒时间,缩短醉酒小鼠醒酒时间。PGP与EGCG协同干预后,可显著改善由高浓度乙醇导致的小鼠胃粘膜损伤,显著减轻小鼠胃组织氧化应激损伤,提高小鼠肝组织ADH及ALDH活力促进乙醇代谢,从而发挥其抗醉解酒作用。 To observe the protective effect of peach gum polysaccharide(PGP)on acute ethanol-induced gastric mucosal injury,and to investigate the anti-inebriation effect and the biological mechanism of peach gum polysaccharide combined with epigallocatechin gallate(EGCG)on mice.The gastric mucosa was simulated by dialysis bag in vitro to study the effect of different concentrations of PGP on the inhibition of ethanol diffusion.In the animal experiment,mice were given 60%edible ethanol for intragastric administration.After different concentrations of EGCG and PGP were given,the mice were observed for the change of drunk tolerance time and hangover time,assessment the gastric mucosal injury,determination of serum ethanol concentration in mice.The contents of alcohol dehydrogenase(ADH)and acetaldehyde dehydrogenase(ALDH)in mouse liver tissues were determined.In vitro and in vivo experiments showed that PGP could significantly inhibit ethanol diffusion,effectively prolong the intoxication time of mice,and shorten the sobering time of drunk mice.The synergistic intervention of PGP and EGCG could significantly improve the gastric mucosal injury induced by high concentration of ethanol in mice,significantly reduce oxidative stress injury of gastric tissue in mice,increase ADH and ALDH activity in liver tissue of mice,and promote ethanol metabolism,thus exerting its anti-drunkenness and anti-alcoholism effect.
