摘要
目的系统总结并全面探讨免疫检查点相关分子与基底样乳腺癌预后的相关性。方法从Gene Expression Omnibus(GEO)数据库下载、合并基于Affymetrix检测平台的乳腺癌全基因组表达谱数据集,并筛选出基底样乳腺癌基因表达谱及预后数据。根据最新文献全面总结免疫检查点基因及免疫检查点抑制剂疗效相关基因,全面分析这些基因的表达在基底样乳腺癌中的预后价值。结果本研究共筛选出618例基底样乳腺癌全基因组表达谱数据及预后信息,并总结了37个免疫检查点基因及免疫检查点抑制剂疗效相关基因。通过全面分析37个免疫检查点相关分子与无进展生存(PFS)的相关性,结果发现,乳酸脱氢酶A(LDHA)与PFS呈负相关(P<0.0001),CD274(程序性死亡蛋白配体1,PDL1)、程序性死亡蛋白1(PDCD1)及细胞毒性T淋巴细胞相关蛋白4(CTLA4)等34个免疫检查点相关分子与PFS呈正相关(P<0.05),乳酸脱氢酶B(LDHB)和YTHDF1与PFS无统计学相关性(P>0.05)。结论37个免疫检查点相关分子中,有34个基因与PFS呈正相关,1个基因与PFS呈负相关,2个基因与PFS无统计学相关性。
Objective To systemically summarize and investigate the prognostic value of immune checkpoint related genes in basal like breast cancer.Methods We downloaded and merged whole genome gene expression profiling data(filtered by Affymetrix platform)of breast cancer from Gene Expression Omnibus(GEO),then choose samples with basal like breast cancer and progression free survival(PFS)data for the study.We Summarized all immune checkpoint molecules and immune checkpoint inhibitor efficacy related genes based on the latest literatures and analyzed their prognostic value in basal like breast cancer.Results A total of 618 basal like breast cancer samples with whole gene expression data and related PFS data were obtained.Among them,we found 37 immune checkpoint related genes through literature mining.Based on survival analysis using gene expression data and PFS data,we demonstrated that lactate dehydrogenase A(LDHA)was negatively correlated with PFS,while 34 genes including CD274(Programmed cell death 1 ligand 1,PDL1),programmed death receptor-1(PD1,PDCD1)and cytotoxic T-lymphocyte-associated protein 4(CTLA4)were all positively correlated with PFS.There were no predictive values of lactate dehydrogenase B(LDHB)and YTHDF1 for PFS.Conclusion Among 37 immune checkpoint related molecules,34 genes are positively associated with PFS,one gene is negatively correlated with PFS,while the rest two genes have no prognostic value.
