摘要
目的采用超声诊断与评价环磷酰胺致小鼠心脏功能的损伤的价值。方法7~8周龄雄性SPF级小鼠18只,随机分为三组:对照组、低剂量组、高剂量组,每组6只大鼠。低剂量组、高剂量小鼠连续腹腔注射环磷酰胺50 mg/(kg·周)、100 mg/(kg·周),对照组小鼠腹腔注射等量0.9%氯化钠溶液,超声评定小鼠的心脏功能损伤情况。结果所有小鼠实验期间都存活,对照组实验前后LVEF、FS、Tei指数、IVRT等比较差异无统计学意义(P>0.05),低剂量组与高剂量组实验后的LVEF、FS低于实验前(P<0.05),Tei指数、IVRT高于实验前(P<0.05),与对照组比较,低剂量组与高剂量组实验后心功能指标变化更显著(P<0.05)。低剂量组与高剂量组实验后实验后dp/dtm和-dp/dtm值比较低于对照组(P<0.05),高剂量组也显著低于低剂量组(P<0.05)。实验后三组的心脏脏器系数比较差异无统计学意义(P>0.05)。结论经超声诊断和评估,环磷酰胺可导致小鼠心脏功能损伤,促使心功能与运动功能下降,且存在剂量依赖性,但不会影响心脏脏器系数。
Objective To evaluate the values of Ultrasound in the diagnosis and evaluation of cyclophosphamide-induced cardiac function damage in mice.Methods 18 male 7-8 weeks old SPF mice were randomly divided into three groups,including control group,low dose group and high dose group,with 6 mice in each group.Low-dose group and high-dose mice were given intraperitoneal injection of cyclophosphamide 50 mg/(kg·week)and 100 mg/(kg·week),the mice in the control group were intraperitoneally injected with 0.9%sodium chloride solution,and the cardiac function damage were evaluated by ultrasound.Results All mice survived during the experiment.There were no significant differences in LVEF,FS,Tei index and IVRT in the control group compared before and after experiment(P>0.05).The LVEF and FS were lower in the low dose group and the high dose group after experiment than those before the experiment(P<0.05),the Tei index and IVRT were higher than those before the experiment(P<0.05),and the difference compared to the control group was statistically significant(P<0.05).The dp/dtm and-dp/dtm values in the low-dose group and the high-dose group were lower than those in the control group(P<0.05),and the high-dose group was also significantly lower than the low-dose group(P<0.05).There were no significant difference in the coefficient of cardiac organ compared among the three groups after experiment(P>0.05).Conclusion By ultrasound diagnosis and evaluation,cyclophosphamide can cause cardiac function damage in mice,which can lead to decrease of cardiac function and motor function with dose-dependent manner,but it does not affect the heart organ coefficient.
