MUC16突变在皮肤恶性黑色素瘤患者预后中的作用分析

Analysis of MUC16 mutation in prognosis of patients with cutaneous malignant melanoma

目的探讨粘蛋白16(MUC16)突变在皮肤恶性黑色素瘤(CMM)临床预后中的作用。方法运用癌症基因组图谱(TCGA)数据库数据集分析MUC16在CMM组织中突变情况,结合GEO数据库数据分析比较MUC16 mRNA在正常皮肤组织、CMM组织中的表达差异,并进一步运用在线数据库分析MUC16突变及其mRNA水平与CMM的预后之间的关系。结果MUC16在CMM中突变频率高达69.3%,其mRNA水平在正常皮肤组织中明显低于CMM组织,差异有统计学意义(P<0.05);生存分析发现MUC16突变的CMM患者具有较好的预后(P<0.05),且MUC16 mRNA表达水平与CMM的预后相关,MUC16表达水平高则预后差(P<0.05)。结论MUC16突变与CMM临床预后密切相关,可以为CMM治疗提供潜在靶点。

Objective To investigate the effect of MUC16 gene in prognosis of cutaneous malignant melanoma(CMM).Methods The Cancer Genome Atlas(TCGA)datasets were used to analyze MUC16 gene mutation in CMM.The GEO database was used to analyze the expression profile of MUC16 in normal skin and CMM tissue.The online databases were used to analyze MUC16 gene mutation and its mRNA level to predict the prognosis of CMM.Results In CMM,the mutation frequency of MUC16 reached 69.3%,and MUC16 mRNA expression was higher in CMM compared with the normal skin tissue(P<0.05).In survival analysis,the cases without MUC16 mutation showed better prognosis(P<0.05),and the expression of MUC16 mRNA was correlated with the prognosis of CMM,the high expression of MUC16 had a poor prognosis(P<0.05).Conclusion MUC16 mutation is correlated to prognosis of CMM,it may provide potential targeting sites to treat the disease.
