大黄素对布鲁菌感染BALB/c小鼠模型的治疗作用 被引量:2

摘要 目的研究大黄素对布鲁菌感染BALB/c小鼠模型的治疗作用,为大黄素抗布鲁菌感染提供实验依据。方法将50只布鲁菌感染小鼠模型随机分为PBS组、多西环素组、大黄素组(2、4、8 mg/mL),第1周每天每只小鼠灌胃1 mL相应药物,第2~3周每3 d灌胃1次。最后1次灌胃结束后检测各组小鼠腹腔积液菌落数、体重、肝脾指数及组织病理变化。结果PBS组、多西环素组、大黄素组(2、4、8 mg/mL)的腹腔载菌量(lg CFU值)分别为5.161±0.501、4.003±0.093、5.175±0.361、4.771±0.498和4.023±0.199;体重分别为(18.047±0.418)g、(18.921±0.291)g、(19.043±0.252)g、(18.816±0.445)g和(19.008±0.430)g;肝脏指数分别为8.913±0.749、6.652±0.748、8.393±0.635、8.074±0.746、7.200±0.466;脾脏指数分别为1.440±0.199、0.554±0.079、0.775±0.120、0.598±0.070、0.546±0.030;与PBS组相比,大黄素组肝脾病理组织有不同程度改善,其中肝脏炎症细胞浸润明显减少,未见变性和坏死,脾脏排列规则有序,红白髓分界清楚,脾小体数量增多。结论大黄素对布鲁菌感染BALB/c小鼠具有一定的抗菌效果,且能够促进肝脾组织结构恢复。 Objective To study the therapeutic effect of emodin on BALB/c mice infected with Brucella and provide experimental basis for treatment of Brucella infection.Methods Fifty mice infected with Brucella were randomly divided into PBS group,doxycycline group,emodin groups(2,4 and 8 mg/ml).Each mouse was given 1 mL of the corresponding drug every day for the first week,and once every 3 days during the second and third weeks.At the end of the last gavage,the therapeutic effect of emodin on mice infected with Brucella were observed through the colony count of ascites,changes of body weight,liver index,spleen index,and pathological changes of liver and spleen stained by HE.Results In PBS group,doxycycline group and emodin groups(2,4 and 8 mg/ml),the intraperitoneal bacterial load(lg CFU)was 5.161±0.501,4.003±0.093,5.175±0.361,4.771±0.498 and 4.023±0.199,respectively;the average weight was(18.047±0.418)g,(18.921±0.291)g,(19.043±0.252)g,(18.816±0.445)g and(19.008±0.430)g,separately;the liver index was 8.913±0.749,6.652±0.748,8.393±0.635,8.074±0.746 and 7.200±0.466,respectively;the index of spleen was 1.440±0.199,0.554±0.079,0.775±0.120,0.598±0.070 and 0.546±0.030,separately.Compared with PBS group,the pathological changes of liver and spleen in emodin groups were improved.In the liver,the infiltration of inflammatory cells was significantly reduced,and degeneration and necrosis was not observed.In the spleen,cells were arranged orderly,the red and white marrow was clearly demarcated,and the number of splenic corpuscles was increased.Conclusion Emodin has certain antibacterial effect on BALB/c mice infected with Brucella.It can also promote the recovery of liver and spleen tissue structure.
