莱菔硫烷对大鼠肾缺血再灌注损伤模型氧化应激的影响 被引量:3

摘要 目的:观察莱菔硫烷(SFN)对肾缺血再灌注损伤(RIRI)大鼠肾组织的形态,功能,H2O2、丙二醛(MDA)含量,超氧化物歧化酶(SOD)活性及其基因表达变化的影响。方法:Wistar大鼠随机分对照组、RIRI组、SFN1组和SFN2组,RIRI组、SFN1组和SFN2组均采用肾动脉钳夹法建立大鼠RIRI模型,SFN1组在夹闭大鼠左侧肾动脉后立即给予莱菔硫烷,SFN2组则在恢复左肾血液灌注时给予莱菔硫烷。缺血再灌注24 h后取血和肾。检测大鼠血清肌酐(SCr)和尿素氮(BUN)含量;观察肾的形态学变化;检测肾组织中H2O2及MDA含量;检测SOD活性及其mRNA和蛋白表达变化。结果:与对照组相比,RIRI组、SFN1组和SFN2组大鼠肾组织形态结构受损明显,血清SCr和BUN含量、肾组织MDA和H2O2含量升高,肾组织SOD活性降低、mRNA和蛋白表达水平升高;SFN1组、SFN2组与RIRI组相比,肾组织形态结构受损轻,血清SCr和BUN含量、肾组织MDA和H2O2含量低,肾组织SOD活性、mRNA和蛋白表达水平高;SFN1组与SFN2组相比,肾组织形态结构损伤较轻,血清SCr和BUN含量、肾组织MDA和H2O2含量较低,肾组织SOD活性较高,SOD mRNA和蛋白表达水平无明显差异。结论:RIRI发生后,莱菔硫烷降低了肾组织的过氧化损伤程度;缺血后即刻给予莱菔硫烷更能有效改善肾的形态结构和功能。 Objective:To observe the effects of sulforaphane(SFN)on histomorphology and function,content of hydrogen peroxide and malondialdehyde(MDA),activity and gene expression of superoxide dismutase(SOD)in the kidney tissue of renal-ischemia-reperfusion-injury(RIRI)rats.Methods:Wistar rats were randomly divided into control group,RIRI group,SFN1 group and SFN2 group.RIRI models of RIRI,SFN1 and SFN2 group were established by clamping the renal artery with artery clamp.The rats of SFN1 group were given sulforaphane immediately after clamping the left renal artery.The rats of SFN2 group were given sulforaphane after restoring left renal blood perfusion.The blood and kidney were taken 24 h after reperfusion.The SCr and BUN content in serum was detected.The morphological changes of the kidney were observed.The MDA and H2O2 content in the kidney were detected;the SOD activity and expression changes of SOD mRNA and protein in the kidney were detected.Results:Compared with the control group,the morphology of kidney tissue of RIRI,SFN1 and SFN2 group was significantly impaired,the SCr and BUN content in serum,MDA and H2O2 content in the kidney were increasd,the SOD activity in the kidney was decreased,the expression level of SOD mRNA and protein were increased.Compared with RIRI group,the damage of morphology of kidney tissues in SFN1 and SFN2 group was lighter,the SCr and BUN content in serum,MDA and H2O2 content in the kidney were lower,the activity,and the expression of mRNA and protein of SOD were increased.Compared with SFN2 group,the damage of morphology of kidney tissues in SFN1 group was lighter,the SCr and BUN content in serum,MDA and H2O2 content in the kidney were lower,the SOD activity in the kidney was higher.There was no significant difference in the mRNA and protein expression level of SOD between the two groups.Conclusion:After RIRI,sulforaphane reduces the degree of peroxidation injury in renal tissues.Immediate administration of sulforaphane after ischemia could more effectively improve the morphology and function of the kidney.
