HPLC同时测定蘑菇中五种核苷类成分含量

摘要 本文利用HPLC对9种蘑菇中胞苷、尿苷、肌苷、鸟苷、腺苷等5种核苷类成分进行含量测定。以水为提取剂,甲醇-水为流动相梯度洗脱,采用Supersil ODS2 C 18色谱柱(5μm,250 mm×4.6 mm),1.0 mL/min流速,259 nm检测波长,30℃柱温,10μL进样量。在该色谱条件下,胞苷、尿苷、肌苷、鸟苷、腺苷在10~100μg/mL范围内线性关系良好,R>0.994。测得9种样品中胞苷、肌苷、鸟苷含量最高为松乳菇,含量分别为159.54 mg/100 g,257.11 mg/100 g,235.79 mg/100 g,尿苷含量最高为杨树蘑,含量为491.24 mg/100g,腺苷含量最高为滑子蘑,含量为207.25 mg/100 g,加标回收率在98.78%~100.57%之间,相对标准偏差在2.31%~3.09%之间。 The content of cytidine,uridine,inosine,guanosine and adenosine in 9 kinds of mushrooms was determined by HPLC in the paper.Nucleosides from samples were extracted by water,gradient elution was performed with methanol and water as flowing phase.The chromatographic conditions were Supersil ODS2 C 18(5μm,250 mm×4.6 mm),flow rate 1.0 mL/min,detection wavelength 259 nm,column temperature 30℃and injection volume 10μL.A good linear relation was showed in the concentration range from 10~100μg/mL with R>0.994 under the chromatographic conditions.Among 9 kinds of samples the content of cytidine,uridine and guanosine from Lactarius deliciosus was the highest and reached at 159.54 mg/100g,257.11 mg/100g and 235.79 mg/100g respectively,the content of uridine from Pholiota cylindracea was the highest and reached at 491.24 mg/100g and the content of adenosine from Pholiota nameko was the highest and reached at 207.25 mg/100g,while the recovery rate of the samples was 98.78%~100.57%with RSD=2.31%~3.09%.
