绿豆药材质量控制方法研究 被引量:2

摘要 为了建立绿豆药材的地方标准,采用经验、显微鉴别法对该药材性状、显微特征进行描述;按照《中华人民共和国药典》2015年版四部通则相关方法,对绿豆中水分、灰分和酸不溶性灰分进行了测定;采用薄层色谱法,以牡荆苷为对照,对药材进行定性鉴别;采用HPLC法,以牡荆苷为对照,采用Agilent ZOR-BAX SB-C18(5μm,4.6 mm×250.0 mm)色谱柱,甲醇-水(40∶60)为流动相,检测波长340 nm下进行定量分析,建立绿豆中牡荆苷的含量测定方法。结果表明,该药材性状、显微鉴别、薄层鉴别方法具有较强的专属性,水分为9.91%~12.57%,总灰分为2.51%~3.22%,酸不溶性灰分为0.03%~0.11%,牡荆苷含量为0.127%~0.248%。上述结果表明,建立的标准具有很好的专属性和准确性,可以作为绿豆药材的地方质量标准。 In order to establish the local quality standards of mung bean,the morphological characters and microscopic characteristics of mung bean were described by means of empirical and microscopic identification.The contents of water,total ash and acid-insoluble ash were analyzed according to the methods of Chinese Pharmacopoeia(2015).Vitexin was used to identify materia medica of mung bean by TLC.HPLC method was used to establish a quantitative method for the determination of vitexin contents in mung bean by using Agilent ZOR-BAX SB-C18(5 mm,4.6 mm×250.0 mm)chromatographic column and methanol-water(40∶60)as mobile phase with detection wavelength of 340 nm.The results showed that the morphological characters,microscopic features and TLC methods were proper,the congtents of water were 9.91%-12.57%,total ash were 2.51%-3.22%,acid-insoluble ash were 0.03%-0.11%,vitexin were 0.127%-0.248%.All of the above data proved that the established quality of coutrol method is specific and accurate,which is suitable for the quality evaluation of mung bean.
