木兰脂素联合5-氟尿嘧啶对人胃癌细胞的影响

摘要 目的探讨木兰脂素联合5-FU对胃癌细胞增殖、凋亡、克隆形成、侵袭性的影响和作用机理。方法用木兰脂素联合5-FU处理胃癌细胞,用MTT法检测细胞增殖,平板克隆实验检测细胞克隆形成能力,用Hochest染色检测细胞凋亡,Transwell检测细胞侵袭能力,比较联合用药与单独给药对胃癌细胞增殖、克隆形成、凋亡、侵袭性的影响。WB检测单独给药和联合用药对胃癌细胞中AKT、ERK信号通路的影响。结果木兰脂素与5-FU联合应用对胃癌细胞增殖的抑制作用较两药单独显著增强,呈协同作用(CI值<1),木兰脂素可以提高5-FU诱导的细胞凋亡、增强对胃癌细胞克隆形成能力、侵袭性的抑制作用。木兰脂素对5-FU的增强作用与AKT、ERK信号通路的抑制有关。结论木兰脂素通过抑制AKT、ERK信号通路增强了5-FU对胃癌细胞的抗肿瘤作用。 Objective To investigate the effects of magolnin combined with 5-FU on proliferation,apoptosis,cloning formation and invasiveness of gastric cancer cells and discover its underlying mechanism.Methods Gastric cancer cells were treated with magolnin combined with 5-FU.MTT assay,plate cloning test,Hoechst staining,transwell assay was used to evaluate the effects of single agents or combined administration on cellular proliferation,cloning ability,apoptosis and invasiveness.Western blotting was used to detect the inhibition of AKT and ERK by single agents or combined administration.Results Combined administration exhibited stronger inhibitory effect on cellular proliferation of gastric cancer cells than magolnin or 5-FU treated alone,showing a synergistic effect(CI<1).Magolnin could increase the apoptosis,enhance the inhibition of colony formation and invasiveness induced by 5-FU,with the blockade of AKT and ERK signaling pathways.Conclusion Magolnin enhances the anti-tumor effect of 5-FU on gastric cancer cells with inhibiting AKT and ERK signaling pathways.
