摘要
目的评价沙棘全果油对昆明种小鼠的免疫调节作用。方法将小鼠适应性喂养到一定体重后随机分为4组,每组40只共160只,按照受试物低、中、高、阴性对照4个不同浓度进行30 d灌胃。其中免疫1组进行巨噬细胞吞噬率和吞噬指数测定试验;免疫2组进行淋巴细胞增殖能力测定和NK细胞活性测定;免疫3组进行溶血空斑数、抗体水平积数测定和足跖肿胀度测定;免疫4组进行碳廓清能力试验。实验结束进行体重和脏器的测定及脏器/体重比值的计算,对各免疫小组的试验数据根据不同灌胃剂量进行方差分析或秩和检验。结果4个免疫组不同灌胃剂量的动物在试验中期、末期的体重相比较差异无统计学意义,脾脏/体重比值和胸腺/体重比值分别与阴性对照组相比,均无显著性差异。单核-巨噬细胞吞噬功能试验、淋巴细胞转化试验、体液免疫试验和碳廓清能力试验结果均为阴性,差异无统计学意义。足趾肿胀试验和NK细胞活性试验结果为阳性。结论沙棘全果油对小鼠具有增强免疫力的功能。
Objective To evaluate the immunomodulatory effect of whole fruit oil of sea buckthorn on kuming mice.Methods After acclimation to a certain body weight,160 mice were randomly divided into four groups and given oral gavage for 30 days.Then,the mice in each of four groups were randomly selected to exposure to four dosages of whole fruit oil of sea buckthorn(i.e.,0、500 ml/kg BW、1000 ml/kg BW、1500 ml/kg BW).The first group was used to measure phagocytosis rate and phagocytosis index,the second group to measure lymphocyte proliferation ability and NK cell activity,the third group to measure the number of hemolytic plaque,the number of antibody levels and the swelling degree of the foot,and the fourth group was used to measure carbon clearance ability.At the end of the experiment,the body weight and viscera were measured and the viscera/body weight ratio was calculated.One-way analysis of covariance or Kruskal-Wallis tests were performed on the data of each of the four groups according to different gavage dosages.Results There was no statistically significant difference in body weight between the four groups with different intragastric dosages in the middle and end of the experiment.Further,no significant differences in spleen/body weight ratio and thymus/body weight ratio were observed.The results of phagocytic function of monocyte macrophage test,lymphocyte transformation test,humoral immunity test and carbon clearance test were all negative,while the results of paw swelling test and NK cell activity test were positive.Conclusion Whole fruit oil of sea buckthorn has the function of enhancing immunity in mice.
