糖通饮与穴位埋线对DPN大鼠血清NGF、TGF-β1的影响 被引量:2

Effects of Tangtong Yin and Acupoint Catgut Embedding on Serum Levels of NGF and TGF-β1 in Rats with Diabetic Peripheral Neuropathy

摘要 目的:探讨糖通饮及穴位埋线治疗糖尿病周围神经病变(DPN)大鼠可能的作用机理及途径,为临床治疗及优选治疗方案提供理论依据。方法:将雄性SD大鼠随机分为正常组、模型组和治疗组(包括穴埋组、中药组和药埋组),通过高能量饲料喂养联合多次小剂量腹腔注射0.1%链脲佐菌霉素的方法建立DPN大鼠模型,造模成功后根据分组进行相应治疗。治疗结束后,检测大鼠血清NGF、血清TGF-β1、FBG、坐骨神经动作电位传导速度及坐骨神经病理形态学变化。结果:治疗后各治疗组大鼠血清NGF含量、坐骨神经动作电位传导速度均较模型组明显升高(P<0.01),各治疗组大鼠FBG、血清TGF-β1含量均较模型组降低(P<0.05或P<0.01),其中药埋组改善较为明显。模型组大鼠坐骨神经形态学较正常组有明显改变,治疗后各治疗组大鼠坐骨神经形态学较模型组有不同程度改善。结论:糖通饮和穴位埋线能通过降低血糖,增加NGF含量,减少TGF-β1含量来达到延缓DPN病程进展的目的,两者合用时效果最佳,值得临床推广应用。 Objective:To explore the possible mechanism and pathway of Tangtong Yin and acupoint catgut embedding in the treatment of diabetic peripheral neuropathy(DPN)through animal experiments,so as to provide theoretical basis and optimal treatment plan for its clinical treatment.Methods:The rats were randomly divided into the normal group,the model group,the treatment groups(the catgut embedding group,the Chinese medication group,the Chinese medicine catgut embedding group).The DPN model was established by high energy diet feeding combined with small dose of intraperitoneal injection of 0.1%STZ solution.Each group was given corrsponding treatment after successful modeling.After 10 weeks of treatment,serum levels of NGF,TGF-β1 and FBG,as well as the changes of sciatic potential conduction velocity and sciatic pathomorphology were detected.Results:After the treatment,the sciatic nerve action potential conduction velocity and serum NGF were significantly increased(P<0.01),the FBG value and serum TGF-β1 were greatly lowered(P<0.05,P<0.01)in the treatment groups compared to those in the model group;of which the Chinese medicine catgut embedding group improved more significantly.Sciatic nerve morphology of the rats was significantly changed in the model group compared to that in the normal group;it was improved in different levels in the treatment groups after the treatment.Conclusion:Tangtong Yin and acupoint catgut embedding can effectively treat DPN and delay the progress of the disease by reducing blood sugar,increasing NGF content,reducing TGF-β1 content,and repairing damaged nerve,which can be achieved better effect when used together.
