加味神术散雾化对FM1感染小鼠NLRP3炎症小体及下游炎性因子表达的影响 被引量:2

Effect of Jiawei Shenshu Powder on the Expression of NLRP3 Inflammatory Bodies and Downstream Inflammatory Factors in FM1-infected Mice

摘要 目的:观察加味神术散雾化对FM1感染小鼠NLRP3炎症小体及下游炎性因子表达的影响。方法:SPF级NIH小鼠80只随机数字分为正常对照组、病毒对照组、利巴韦林组以及神术散雾化组各20只。对正常对照组外的各组予以复制流感病毒FM1感染小鼠模型,造模前2日开始给药及给药第3日病毒接种1h后继续给药。动物给药剂量按动物每公斤体质量占人体表面积的比值计算,正常组和病毒对照组使用频率和剂量同神术散雾化组。连续给药7日,比较各组小鼠咽部黏膜NLRP3、caspase-1的mRNA表达、咽部黏膜匀浆IL-1β、IL-18的分泌水平。结果:与正常对照组比较,病毒对照组、利巴韦林组及神术散雾化组NLRP3、caspase-1的mRNA相对表达量、IL-1β、IL-18水平较高(P<0.05);与病毒对照组比较,利巴韦林组及神术散雾化组NLRP3、caspase-1的mRNA相对表达量、IL-1β、IL-18水平较低(P<0.05);与利巴韦林组比较,神术散雾化组NLRP3、caspase-1的mRNA相对表达量、IL-1β、IL-18水平较低(P<0.05)。结论:加味神术散雾化能够下调FM1感染小鼠NLRP3炎性小体及其下游因子IL-1β、IL-18的分泌水平,可能对其产生免疫保护机制。 Objective:To investigate the effect of Jiawei Shenzhu Powder on the expression of NLRP3 inflammatory bodies and downstream inflammatory factors in FM1-infected mice.Method:88 SPF-class NIH mice were divided into normal control group,virus control group,ribavirin group and Shenzhu Powder atomization group evenly in random.A mouse model of influenza virus FM1 infection was replicated in each group except the normal control group and administration was continued on the 3rd day before the model establishment and administration on the 3rd day after the virus was inoculated for 1 hour.The dose of the animal was calculated according to the ratio of the body surface area per kilogram of body weight of the animal.The frequency and dose of the normal group and the virus control group were the same as the Shenzhu Powder atomization group,with continuous administration for 7 days,The mRNA expressions of NLRP3 and caspase-1 in pharyngeal mucosa of each group were compared.The levels of IL-1βand IL-18 in pharyngeal mucosa were compared,too.Result:Compared with that of the normal control group,the relative expressions of NLRP3 and caspase-1 mRNA,IL-1βand IL-18 were higher in the virus control group,ribavirin group and Shenzhu Powder group(P<0.05).Compared with that of the virus control group,the relative expression of NLRP3 and caspase-1 mRNA,IL-1βand IL-18 was lower in the ribavirin group and Shenzhu Powder group(P<0.05).Compared with that of the ribavirin group,the mRNA expression level of NLRP3 and caspase-1 and the level of IL-1βand IL-18 in Shenzhu Powder group was lower(P<0.05).Conclusion:Modified Shenzhu Powder could down-regulate the secretion levels of NLRP3 inflammatory corpuscles and its downstream factors IL-1βand IL-18,which probably have immunoprotective effect on mice infected with influenza virus FM1.
